中文摘要: 双孢菇菇柄中含有丰富的核酸,从菇柄中提取核酸物质,为菇柄的后续处理提供了新的途径。研究微波法提取 新鲜双孢菇菇柄中核酸的得率和色差值的影响因素,在单因素的试验基础上,通过正交试验优化了核酸的提取工艺参 数。得到提取工艺的最佳条件为:微波强度 16 W/g,微波时间 40 s,菇柄厚度 14 mm,氯化钠浓度 0.9 %,在此条件下核酸 得率为 3.24 %,色差值为 75.10。与传统的氨法、浓盐法相比,核酸得率分别增加了 10.96 %、58.82 %,且所得核酸色泽 更好。

Abstract:Agaricus bisporus stipe is rich in nucleic acid. Extracting nucleic acid from stipe will be a new way on subsequent processing about stipe.In this paper, extraction of nucleic acid yield and color value in Agaricus bisporus stipe by microwave technology was studied, on the basis of single factor experiments, using orthogonal test optimizes the extraction of nucleic acid process parameters. the optimal conditions for extraction was ob- tained: microwave intensity 16 W/g, microwave time 40 s, stipe thickness 14 mm, sodium chloride concentra- tion 0.9 %. Under these optimum conditions, Nucleic acid yield was 3.24 %, and L* value was 75.10. Com- pared with the traditional ammonium method and strong salt method, the yield was increased by 10.96 % and 58.82 % respectively, in addition, microwave extraction had a better color.
