中文摘要: 应用甲基噻唑基四唑(MTT)比色法检测普洱茶水提物对 6 种不同肿瘤细胞株生长抑制作用以及流式细胞仪分 析普洱茶水提物对 Hela 细胞凋亡的影响;采用裸鼠移植瘤模型,观察普洱茶水提物对宫颈癌的治疗作用。体外细胞毒 活性检测显示,普洱茶水提物对人肝癌 BEL-7402、胃癌 MGC-823、乳腺癌 MCF-7、子宫颈癌 HeLa 及结肠癌 HT-29 均 有抗肿瘤作用,其中普洱茶水提物 200 μg/mL 浓度下肿瘤细胞生长抑制率可达 79 %以上。普洱茶水提物能诱导 Hela 细 胞发生凋亡,当浓度为 100 μg/mL,细胞凋亡率为(50.92±5.38)%。荷瘤鼠实验结果显示:连续给药 28 d,普洱茶水提物1.0 g/kg 剂量组、0.5 g/kg 剂量组对人宫颈癌 Hela 移植瘤抑制率分别为 40.3 %、22.4 %。普洱茶水提物在体外和体内均具 有一定的抗肿瘤作用。
Abstract:The aqueous extract of Pu'er anti-tumor effect was tested by growth inhibition of six different tumor cell lines application of methyl thiazolyl tetrazolium (MTT) colorimetric assay and its apoptosis inducing effect was analyzed by Flow cytometry in Hela cell. The aqueous extract of Pu'er anti-tumor effect was also tested in mouse xenograft model on the cervical cancer. In vitro data showed that the aqueous extract of Pu'er could in- hibit BEL-7402,MGC-823,MCF-7,Hela and HT-29 cell line. The tumor inhibition rate could reach as high as 79 % in the concentration of 200 μg/mL.The Flow cytometry results showed that the apoptosis rate can be in- duced as high as(50.92±5.38) % in the concentration of 100 μg/mL. In vivo data showed that after administra- tion of 28 days, 1.0 g/kg dose group、0.5 g/kg dose group of aqueous extract of Pu'er extract on human cervical cancer Hela transplanted tumor inhibition rates were 40.3 % and 22.4 %. Aqueous extract of Pu'er has the effect of antitumor in vitro and in vivo and a certain dose-effect relationship.
