中文摘要: 以乙醇-水为提取剂,总黄酮提取量为考察指标,通过醇提取法提取当归中的总黄酮成分。通过单因素试验分 别考察乙醇体积分数、料液比、提取时间、提取温度对总黄酮提取量的影响,在此基础上利用 Design-Expert 软件 Box-Behnken 中心组合设计法设计响应面分析料液比、乙醇体积分数、提取时间和提取温度 4 个因素对总黄酮提取量的影 响,并通过方差分析回归建立数学模型。料液比 1 ∶ 11(g/mL),乙醇体积分数 70 %,提取时间 2.5 h,提取温度 60 ℃,在此 条件下,总黄酮提取量达到 0.022 8 mg/g。结果表明:Box-Behnken 设计结合响应面分析法可以很好地对当归总黄酮提取 工艺进行优化。

Abstract:Using ethanol- water as extracting agent, total flavonoids extraction amount as the indexes, through the ethanol extraction method to extract from Angelica sinensis total flavonoids. The effects of ethanol volume fraction, ratio of material to liquid, extraction time and extraction temperature were investigated by single factor experiment. The Design-Expert software Box-Behnken center combination design method was used to design response surface analysis. The statistical analysis and mathematical modeling were also applied for analysis the data. The results showed that the optimal extraction parameters would be the material liquid ratio of 1 ∶ 11(g/mL), ethanol volume fraction of 70 % , extraction time of 2.5 h, and extraction temperature of 60 ℃ . Under these conditions, total flavonoids extraction amount reached 0.022 8 mg/g. Box-Behnken design combined with response surface analysis method can be used to optimize the extraction process of total flavonoids from Angelica sinensis.
