中文摘要: 研究龙眼壳粗黄酮提取物体内外抗肿瘤作用。 体外试验以 MTT 法检测龙眼壳粗黄酮提取物对食管癌细胞 EC109、肝癌细胞 HepG2 的增殖抑制作用。体内试验建立 S180 荷瘤小鼠模型,观察龙眼壳粗黄酮提取物(100、200、 300 mg/kg)组和龙眼壳粗黄酮提取物+CTX(20 mg/kg)组的抑瘤率,计算提取物+CTX 组小鼠的脾脏指数、胸腺指数、巨噬 细胞吞噬指数、吞噬率,并采用 ELISA 法检测提取物组小鼠血清 TNF-α 的水平。体外试验显示龙眼壳粗黄酮提取物可 显著抑制 EC109、HepG2 细胞的生长,并呈现一定的浓度依赖性。体内试验显示龙眼壳粗黄酮提取物(低)、(中)、(高)组 均可抑制 S180 肉瘤生长,抑瘤率分别为 28.24 %、44.91 %、60.65 %;提取物(低)+CTX 组、提取物(中)+CTX 组、提取物(高)+CTX 组抑瘤率分别为 53.7 %、65.74 %、70.37 %,均高于 CTX 组 47.69 %,且与 CTX 组比较,提取物+CTX 组可不同 程度地升高荷瘤小鼠的脾脏指数、胸腺指数、巨噬细胞的吞噬率、吞噬指数;和模型组比较,提取物(低)、(中)、(高)组不 增加小鼠血清 TNF-α 水平。龙眼壳粗黄酮提取物可抑制肿瘤细胞生长,具有抗肿瘤作用,且对 CTX 化疗药物具有增效 减毒作用。

Abstract:To investigate the antitumor effects of coarse flavones extract from longan shell in vitro and vivo. In vitro, MTT method was used to detect the inhibitory effect of coarse flavones extract from longan shell on EC109 and HepG2. In vivo, S180-bearing mice were given coarse flavones extract from longan shell and the tumor inhibitory rates of mice were detected in coarse flavones extract (100,200,300 mg/kg) groups and coarse flavones extract combined with CTX (20 mg/kg) groups. The thymus index, spleen index, macrophage phagocytic rate and phagocytic index were calculated in coarse flavones extract combined with CTX proups. ELISA method was used to detect the the levels of serum TNF-α in coarse flavones extract groups. In vitro, the results showed that the coarse flavones extract of longan shell significantly inhibited the growth of EC109 and HepG2 cells, and showed a certain concentration dependence.In vivo, the results showed that the coarse flavones extract of longan shell (low), (middle) and (high) group could inhibit the growth of S180 sarcoma, and the inhibition rate was 28.24 %, 44.91 % and 60.65 % respectively. The tumor inhibitory rates of coarse flavones extract combined with CTX were 53.7 % ,65.74 % ,70.37 % , which were higher than that of CTX administration alone (47.69 % ). Compared with those in CTX group,the thymus index, spleen index, macrophage phagocytic rate and phagocytic index were significantly elevated in all coarse flavones extract combined with CTX groups. Compared with the model group, the extract (low), (middle) and (high) groups did not increase the level of serum TNF-αin mice. The coarse flavones extract from exocarp of longan had antitumor effect and could inhibit the growth of S180 tumor and and had synergistic attenuation effect of CTX.
