中文摘要: 采用单因素试验和正交试验优化了杨树桑黄摇瓶产胞外多糖(EPS)的培养条件,并研究了优化培养基中菌丝 体的形态变化。单因素试验结果表明,杨树桑黄产 EPS 的最适生长周期为 8 d,最佳碳源、氮源和无机盐分别为蔗糖、玉 米粉和 KH2PO4,最适环境条件为温度 28 ℃,pH 7。在此基础上,选用碳源、氮源和无机盐 3 个因素,应用正交试验优化出 杨黄产 EPS 的最佳培养基配方为:40 g/L 蔗糖,4 g/L 玉米粉,4 mmol/L KH2PO4。 在优化培养条件下, 胞外多糖产量为0.352 g/L。优化培养基的形态学结果表明,随着发酵时间的延长,菌丝球逐渐增大,平均直径、紧密度均逐渐增加,圆度先 增加后急剧减小,粗糙度与圆度呈负相关,表现为先减小后增加的趋势。

Abstract:In this study, the single-factor method and orthogonal design were used to optimize the flask culture conditions for exopolysaccharide (EPS) production by Phellinus vaninii Ljup. The morphological changes in optimal medium mycelia were also investigated. The results of single -factor experiment for EPS production showed the optimum growth cycle 8 days. And the best carbon source, nitrogen source and inorganic salts were sucrose, corn steep powder and KH2PO4, the optimal temperature of 28 ℃, pH 7. Then, the concentrations of three factors, i.e. carbon source, nitrogen source and inorganic salts was optimized using the orthogonal design and the optimum culture medium for EPS production were 40 g/L sucrose, 4 g/L corn steep powder, 4 mmol/L KH2PO4. Under the optimial culture conditions, EPS yield was 0.352 g/L. The morphology under the optimal medium was invesitaged during the fermentation period. The mycelial pellets increased gradually, and the average diameter and density also increased, but roundness increased first then decreases sharply. The roughness first decreased and then increased, which was negatively correlated with roundness.
