中文摘要: 研究祁州漏芦提取物对 D-氨基半乳糖(GalN)诱发的小鼠急性肝损伤的保护作用。将小鼠随机分为正常组、模 型组、联苯双脂组、祁州漏芦乙醇提取物组、祁州漏芦正丁醇层萃取物组和水层萃取物组。每日给药 1 次,连续 7 d。实验 末期,腹腔注射 GalN 建立急性肝损伤模型。比色法检测血清谷丙转氨酶(ALT)、谷草转氨酶(AST)以及肝脏诱导型一氧 化氮合酶(iNOS)、一氧化氮(NO)、丙二醛(MDA)、还原型谷胱甘肽(GSH)、谷胱甘肽过氧化物酶(GPx)、超氧化物歧化酶(SOD)水平。祁州漏芦不同溶剂层提取物显著降低 GalN 致急性肝损伤小鼠血清 ALT、AST 活性(P<0.05);降低肝组织 iNOS 和 NO 水平,降低肝线粒体 MDA 水平(P<0.05);升高肝线粒体 GPx、SOD 活性和 GSH 水平(P<0.05)。祁州漏芦提取 物对 GalN 致急性肝损伤具有保护作用,其机制可能与其抗氧化和抗炎作用有关。
Abstract:To investigate the protective effect of Rhaponticum uniflorum extracts on liver injury induced by D-galactoamine(GalN) in mice, mice were randomly assigned to the normal control, model, bifendate, Rhaponticum uniflorum ethanol extract, as well as the butanol (RUBF) and aqueous fractions of Rhaponticum uniflorum. Animals were treated once daily for 7 days. At the end of the experiment, GalN was given intraperitoneally to the mice of groups, then the alanine aminotransferase (ALT), aspartate aminotransferase (AST), inducible nitric oxide synthase (iNOS), nitric oxide (NO), malondialdehyde (MDA), glutathione (GSH), glutathione peroxidase (GPx) and superoxide dismutase (SOD) were detected with the colorimetric method. The results showed that the administration with different solvent fractions of Rhaponticum uniflorum reduced the serum activities of ALT and AST (P<0.05), decreased hepatic iNOS and NO level, decreased the MDA content and increased GPx, SOD and GSH levels of liver mitochondria in mice with acute liver injury (P<0.05). This study showed that Rhaponticum uniflorum extracts had protective effects on acute liver injury induced by GalN in mice, probably via anti-oxidative and anti-inflammatory activities.
