中文摘要: 观察丝瓜提取物(Luffa extract,LE)对 β-淀粉样蛋白(Aβ25-35)诱导的 PC12 细胞损伤的保护作用。应用 Aβ25-35 造 成神经细胞损伤模型,用四甲基偶氮唑盐微量酶反应(MTT)比色法检测各剂量组丝瓜提取物对 PC12 细胞活力的变化, 检测过氧化氢酶(CAT)、谷胱甘肽过氧化氢酶(GSH-Px)活性水平,通过蛋白印迹试验(Western Blotting)来检测 B 细胞 淋巴瘤基因-2(Bcl-2)和 Bcl-2 相关 X 蛋白(Bax)的表达水平。丝瓜提取物剂量组 PC12 细胞存活率高于模型组,GSHPx 活性显著增强,CAT 含量显著增加(P<0.01 或 P<0.05),Bax 表达水平降低,Bcl-2 表达增高,与模型组比较,差异显著(P<0.05~P<0.01)。丝瓜提取物对 Aβ25-35 诱导的 PC12 细胞损伤有明显的保护作用。
Abstract:To study the protective effect of luffa extract on injury of PC12 cells induced by amyloid β protein (Aβ25-35). Neurons injury model was induced by Aβ25-35. MTT assay was used to detect the cellular viability after different concentrations of luffa extract. The activity of catalase (CAT) and glutathione peroxidase (GSH-Px) were detected. The expression of Bcl-2 and Bax were detected by Western Blotting. Compared with the control group, the cellular viability of luffa extract group was increased, the activity of GSH -Px and CAT were significantly increased (P<0.01, P<0.05), the expression of Bax was decreased and Bcl-2 was increased (P<0.05-P<0.01). luffa extract can protect PC12 cells from Aβ25-35 induced cytotoxicity.
