中文摘要: 该研究通过水蒸气蒸馏法提取甜罗勒精油,采用GC-MS进行成分分析并初步探究其体外抗氧化活性;以甜罗勒精油(EO)为芯材,以壳聚糖(CS)为壁材,通过离子交联法制备得到负载甜罗勒精油的壳聚糖纳米粒(CSNPs-EO),通过响应面优化法获得该CSNPs-EO最佳制备条件并对CSNPs-EO进行形貌表征。实验结果表明EO主要成分为芳樟醇(21.16%)、丁香酚(20.83%)、τ-杜松醇(8.63%)和桉叶油素(6.60%)等萜类和苯丙素衍生物,对DPPH自由基、ABTS自由基均具有良好的清除作用,其IC50值分别为0.407 mg·mL?1 和0.355 mg·mL?1 ;CSNPs-EO最佳制备条件为壳聚糖溶液pH 4.70,芯壁比1.0,TPP溶液浓度2.95%,此条件下包埋率为71.14 %,载药率为41.56%;通过TEM和DLS分析,CSNPs-EO为近圆球型核壳结构,平均粒径为178.1 nm,多分散指数(PDI)为0.163,Zeta电位为39.1mV。甜罗勒精油具有良好的抗氧化活性,通过离子交联法制备的CSNPs-EO粒径较为均一,稳定性良好。
Abstract:IIn this study, essential oil was extracted from sweet basil through steam distillation, its components were analyzed by GC-MS, and its antioxidant activity was preliminarily explored in vitro. Utilizing sweet basil essential oil (EO) as the core material and chitosan (CS) as the wall material, chitosan nanoparticles loaded with sweet basil essential oil (CSNPs-EO) were fabricated via ionic crosslinking. The optimal preparation conditions for CSNPs-EO were ascertained using response surface methodology, and the morphological characteristics of CSNPs-EO were delineated. The experimental findings revealed that the principal constituents of EO were terpenes and phenylpropanoid derivatives, encompassing linalool (21.16%), eugenol (20.83%), τ-cadinol (8.63%), and cineole (6.60%). EO demonstrated potent scavenging capabilities against both DPPH and ABTS radicals, with IC50 values of 0.407 mg·mL?1 and 0.355 mg·mL?1, respectively. The optimal preparation parameters for CSNPs-EO included a chitosan solution pH of 4.70, a core-to-wall ratio of 1.0, and a TPP solution concentration of 2.95%. Under these conditions, the embedding efficiency reached 71.14%, and the drug loading capacity was 41.56%. TEM and DLS analyses indicated that CSNPs-EO possessed a nearly spherical core-shell architecture, with an average particle diameter of 178.1 nm, a polydispersity index (PDI) of 0.163, and a zeta potential of 39.1 mV. Sweet basil essential oil exhibits remarkable antioxidant activity, and CSNPs-EO prepared through ionic crosslinking display uniform particle size distribution and favorable stability.
