中文摘要: 炎症性肠病(inflammatory bowel disease,IBD)发病率持续攀升,但传统治疗方式存在一定局限,基于益生菌及其发酵乳制品的干预策略成为研究热点。该文采用副干酪乳酪乳杆菌Jlu-CH9与商业发酵剂共发酵制备酸奶,对该酸奶在储藏期间的持水力、质构特性及流变学特性等进行检测,并评估该酸奶对葡聚糖硫酸钠(dextran sulfate sodium,DSS)诱导的小鼠结肠炎的缓解作用。结果表明Jlu-CH9联合发酵可显著改善酸奶品质,表现为酸奶持水力提高,硬度、内聚性、胶着性及胶黏性等质构特性增强,储能模量增加,凝胶网络结构更加稳定。动物实验结果表明,联合发酵酸奶能够显著缓解DSS诱导的结肠炎的症状,包括小鼠体重下降、疾病活动指数(disease activity index,DAI)评分升高及结肠缩短,减轻结肠组织病理损伤,降低肠道通透性,上调E-cadherin、Occludin和ZO-1等紧密连接蛋白表达,下调TNF-α、IL-1β、IL-6和IL-17A等促炎因子表达,提高IL-10等抗炎因子表达水平。综上,Jlu-CH9作为辅助发酵剂能够改善酸奶品质,又能赋予其保护肠道屏障、缓解结肠炎的功能特性,在IBD的改善中有良好的应用前景。
Abstract:To investigate the effect of Lacticaseibacillus?paracasei (Jlu-CH9) as an auxiliary starter culture on yogurt quality, and to evaluate its ability to alleviate dextran sulfate sodium (DSS)-induced colitis in mice. Yogurt was prepared by combining Jlu-CH9 with a commercial starter culture, and the pH, titratable acidity, water-holding capacity, texture, and rheological properties of the yogurt were measured during storage. An acute colitis model induced by DSS in mice was established, and the intervention effects of the yogurt containing the combined starter culture on colitis were systematically evaluated by monitoring the mice’s body weight, Disease Activity Index(DAI), colon length, histopathological changes, intestinal permeability, expression of tight junction proteins in the intestinal mucosa, and levels of inflammatory factors to systematically evaluate the intervention effects of co-fermented yogurt on colitis. The results showed that, compared with fermentation using commercial starter cultures alone, co-fermentation with Jlu-CH9 significantly improved yogurt quality, as evidenced by increased water-holding capacity, enhanced textural properties such as hardness, cohesiveness, adhesiveness, and viscosity, an increase in the storage modulus, and a more stable gel network structure. Results from animal experiments indicated that co-fermented yogurt significantly alleviated DSS-induced weight loss, elevated DAI scores, and colon shortening in mice; reduced histopathological damage to the colon; lowered intestinal permeability; upregulated the expression of tight junction proteins such as E-cadherin、Occludin and ZO-1, downregulate the expression of pro-inflammatory factors such as TNF-α, IL-1β, IL-6, and IL-17A, and increase the levels of anti-inflammatory factors such as IL-10. Therefore, it can be concluded that Jlu-CH9, as a co-fermentation starter, improves yogurt quality and confers beneficial intestinal protection. Yogurt fermented with this strain alleviates DSS-induced colitis in mice by enhancing intestinal mucosal barrier function and regulating inflammatory responses, thereby providing a theoretical basis for the development of functional probiotic yogurt and its application in the field of intestinal health.
