中文摘要: 以舟山秋茄果实为原料,采用响应面法优化超声辅助提取多糖的工艺,并通过高效液相色谱、傅里叶变换红外光谱对多糖进行结构表征,分析其单糖组成、分子量和官能团特征。通过DPPH自由基、ABTS+自由基清除试验以及Fe3+还原试验评价其体外抗氧化活性,并应用CCK-8法检测其对小鼠乳腺癌4T1细胞和EMT6细胞的增殖抑制作用。结果表明,秋茄果实多糖最佳提取工艺参数为超声温度71 ℃、液料比20∶1 (mL/g)、超声时间94 min,在此条件下多糖提取率为20.809%,总糖含量为71.771%。结构表征结果显示,该多糖由葡萄糖、半乳糖醛酸、半乳糖和阿拉伯糖组成(物质的量比为106.0∶3.4∶1.1∶1.0),主要分子量为295.249 kDa,是一种含有α-糖苷键、β-糖苷键的酸性吡喃多糖。活性研究表明,该多糖具有显著的抗氧化和抗肿瘤活性,其对DPPH自由基、ABTS+自由基的清除率和Fe3+的相对还原力的半数有效浓度(half maximal effective concentration,EC50)分别为0.028、0.314、1.172 mg/mL,对4T1细胞和EMT6细胞的半数抑制浓度(half maximal inhibitory concentration,IC50)分别为0.593 mg/mL和0.681 mg/mL。
Abstract:This study utilized Kandelia candel fruits from Zhoushan as the raw material to extract polysaccharide and employed response surface methodology to optimize the ultrasound-assisted extraction process. The polysaccharide was structurally characterized in terms of monosaccharide composition, molecular weight, and functional groups via high-performance liquid chromatography and Fourier transform infrared spectroscopy.The in vitro antioxidant activity of the extracted polysaccharide was evaluated through DPPH radical scavenging, ABTS+ radical scavenging, and Fe3+ reduction assays. The inhibitory effects of the polysaccharide on the proliferation of mouse mammary carcinoma 4T1 and EMT6 cells was assessed via the CCK-8 assay. The results indicated that the optimal extraction parameters for polysaccharide from K. candel fruits were ultrasonication temperature of 71 ℃, a liquid-to-solid ratio of 20∶1 (mL/g), and ultrasonication time of 94 min. Under these conditions, the polysaccharide yield reached 20.809%, with the total sugar content of 71.771%. Structural characterization revealed that the polysaccharide comprised glucose, galacturonic acid, galactose, and arabinose (at a molecular ratio of 106.0∶3.4∶1.1∶1.0), with a primary molecular weight of 295.249 kDa. It was identified as an acidic pyranose polysaccharide containing α,β-glycosidic bonds. Bioactivity studies demonstrated significant antioxidant and antitumor activities. The polysaccharide exhibited EC50 values of 0.028, 0.314, and 1.172 mg/mL for scavenging rates against DPPH radicals, ABTS+ radicals, and for the relative reducing power of Fe3+, respectively. Its IC50 values against 4T1 and EMT6 cells were 0.593 mg/mL and 0.681 mg/mL, respectively.
