中文摘要: 按照玉米胚芽水酶法提油工艺,将玉米胚芽浸泡于柠檬酸缓冲液中,热处理后沥水。沥水后的玉米胚芽破碎、酶解后,玉米胚芽中蛋白质主要分为两部分:一部分存在于热处理的浸泡液中,另一部分存在于酶解液中。将浸泡液和酶解液分别浓缩、冷冻干燥,提取浸泡液和酶解液中的可溶性蛋白质,以研究玉米胚芽在不同热处理条件下,浸泡液及酶解液中蛋白质的溶出率、乳化能力及可溶性蛋白质分子量分布。结果表明:浸泡液在pH3~5、热处理时间30~90 min条件下,pH值越低,热处理时间越长,浸泡液和酶解液中蛋白质溶出率越高。浸泡液pH值及热处理时间对酶解液中蛋白质的乳化能力和可溶性蛋白质分子量分布影响较大,而对浸泡液中蛋白质的乳化能力和可溶性蛋白质分子量分布影响很小。与酶解液中可溶性蛋白质相比,浸泡液中的可溶性蛋白质主要是低相对分子量蛋白质,其乳化能力较低,对清油提取率影响小。
Abstract:According to the process of aqueous enzymatic extraction(AEE),the soluble protein in corn germ was divided into two parts after corn germ soaking in citric acid buffer,hydrothermal treatment (HTT),draining,crushing,and enzymolysis. One part was in the soaking solution,and the other part was in the enzymatic hydrolysate. The soaking solution and the enzymatic hydrolysate were separately concentrated and freeze-dried to prepare the soluble protein. The influences of buffer pH and HTT time on the yield,emulsifying capacity(EC) of the protein and molecular weight distribution (MWD) of the soluble protein in the soaking solution and the enzymatic hydrolysate were studied. The results showed that lower buffer pH within the range of pH3-5 and longer HTT time within the range of 30-90 min were conductive to the yield improvement of protein in the soaking solution and enzymatic hydrolysate. Buffer pH and HTT time had stronger influences on EC of the protein and MWD of the soluble protein in the enzymatic hydrolysate and little influences on EC of the protein and MWD of the soluble protein in the soaking solution. The soluble protein in the soaking solution had lower relative molecular weight than that in the enzymatic hydrolysate,which showed that it had little influence on the yield of free oil.
