紫甘薯浆酶解工艺优化及其挥发性风味物质分析

中文摘要: 以‘徐紫薯8号’为研究对象,对其低温α-淀粉酶酶解工艺进行优化,并基于气相色谱-离子迁移谱(gas chromatography-ion mobility spectrometry,GC-IMS)联用技术分析加热和酶解处理前后紫甘薯浆的挥发性风味物质的变化。结果表明:低温α-淀粉酶酶解紫甘薯浆的最优工艺为pH6.0、酶添加量40 U/g、酶解温度 45 ℃、酶解时间60 min,酶解后紫甘薯浆中的还原糖含量为1.77%。GC-IMS通过差异对比图和指纹图谱均可直观地区分不同孵育温度和酶解前后的紫甘薯浆样品。紫甘薯浆在40 ℃和85 ℃下分别鉴定出58种和73种香气成分,加热有利于紫甘薯浆中挥发性风味物质的释放。酶解处理可以增加紫甘薯浆中醛类、醇类、酮类和酯类等51种挥发性风味物质的浓度,表现为典型的脂香、花香、果香、酒香等气味特征。主成分分析(principle component analysis,PCA)结果显示,PC1、PC2贡献率分别为85%和10%,累计贡献率达到95%。
Abstract:The cold-adapted α-amylase enzymatic hydrolysis process of 'Xuzishu No. 8' purple sweet potato was optimized. Gas chromatography-ion mobility spectrometry(GC-IMS) technology was used to analyze the changes in volatile flavor components in purple sweet potato pulp before and after heating and enzymatic hydrolysis. The results showed that the optimal hydrolysis conditions of purple sweet potato pulp with the α-amylase were as follows:pH of 6.0,enzyme amount of 40 U/g,enzymatic hydrolysis temperature of 45 ℃,and enzymatic hydrolysis time of 60 min. Under the optimal enzymatic hydrolysis conditions,the reducing sugar content of purple sweet potato pulp was 1.77%. Purple sweet potato pulp samples at different incubation temperatures and before and after enzymatic hydrolysis could be visually distinguished by the difference comparison map and fingerprint of GC-IMS. In total,58 and 73 aroma components were identified in purple sweet potato at 40 ℃ and 85 ℃,respectively. Heating was beneficial to the release of volatile flavor components in purple sweet potato pulp. Enzymatic hydrolysis could increase the concentration of 51 volatile flavor components such as aldehydes,alcohols,ketones,and esters in purple sweet potato pulp,which showed typical odor characteristics such as lipid,floral,fruity,and wine. Principle component analysis (PCA) showed that the contribution rate of PC1 and PC2 was 85% and 10%,respectively,and the cumulative contribution rate reached 95%.
