中文摘要: 为探究低压静电场技术对半滑舌鳎微冻贮藏过程中品质的影响,通过对半滑舌鳎的鲜度指标与蛋白质特性指标进行分析,探究不同(0、2 000、2 500 V/m)低压静电场处理对微冻贮藏半滑舌鳎肌肉品质的影响。结果表明,随着贮藏时间的延长,各组样品肌肉中肌原纤维蛋白含量、钙离子依赖性腺苷三磷酸酶(calcium-dependent adenosine triphosphatase,Ca2+-ATPase)活性、总巯基含量和溶解度均呈下降趋势且低压静电场组对于上述指标保持相对较好;各组样品蛋白质表面疏水性、羰基含量和蛋白质溶解性呈上升趋势,且对照组样品明显高于低压静电场组;通过电泳和微观结构结果显示,贮藏至第30天时,各组样品肌肉中蛋白质均出现降解现象,且2 500 V/m低压静电场组贮藏的样品肌肉中蛋白质降解程度明显低于其他两组;通过微观结构观察发现,贮藏至第30天时低压静电场组样品肌肉结构相对较好。综上,低压静电场技术可延缓半滑舌鳎肌肉品质的劣化。
Abstract:This study aimed to explore the effect of low-voltage electrostatic field technology on the quality of Cynoglossus semilaevis during micro-freezing storage. By analyzing the freshness index and protein characteristic index of Cynoglossus semilaevis,the study investigated the effect of different (0,2 000,and 2 500 V/m) low voltage electrostatic field treatment on the muscle quality of Cynoglossus semilaevis. The results indicated that,with the extension of storage time,the myofibrillar protein content,calcium-dependent adenosine triphosphatase(Ca2+-ATPase) activity,total sulfhydryl content,and solubility showed a decreasing trend in muscle samples of all groups,while the low-voltage electrostatic field group demonstrated relatively ideal preservation of these indices. Conversely,the protein surface hydrophobicity,carbonyl content,and protein solubility exhibited an increasing trend in all groups,with the above three items being significantly higher in the control group.Sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) results showed that at 30 days of storage,protein degradation occurred in the muscle of all groups,with the degree of protein degradation being significantly lower in the muscle of the 2 500 V/m low-voltage electrostatic field group. Microstructure observation of muscle showed that at 30 days of storage,the muscle structure of the low-voltage electrostatic field group samples was relatively ideal. In conclusion,the low-voltage electrostatic field technology could delay the muscle quality deterioration of Cynoglossus semilaevis.
