茯苓对小鼠肠道菌群的影响

中文摘要: 为研究不同提取方法的茯苓对小鼠调节肠道菌群的影响,该实验将130 只昆明小鼠随机分为阴性对照组,生茯苓粉低、高剂量组,熟茯苓粉低、高剂量组,茯苓代培粉低、高剂量组,茯苓水提物低、中、高剂量组,茯苓醇提物低、中、高剂量组,连续灌胃给药7 d。末次给药后,收集小鼠结肠段粪便进行肠道菌群16S rDNA 检测。结果表明:不同方法提取的茯苓主要功能包括新陈代谢、遗传信息加工、环境信息处理、细胞转化和人类疾病相关基因等。然而,与阴性对照组比较,茯苓醇提物组的α 多样性指数最高,且茯苓醇提物高剂量组的操作分类单元(operational taxonomic units,OTUs)菌群种类最多,其中变形菌门(Proteobacteria)中的毛螺旋菌属(Parasutterella)丰度最高;生茯苓粉低剂量组疣微菌门(Verrucomicrobiota)中的阿克曼菌属(Akkermansia)丰度最高。不同提取方式的茯苓均可以调节小鼠的肠道菌群,并对肥胖及相关代谢性疾病有改善作用。

Abstract:To investigate the effect of different extraction methods of Poria cocos on the regulation of intestinal flora in mice, 130 KM mice were randomly divided into a negative control group, low-dose and high-dose groups of raw P. cocos powder, low-dose and high-dose groups of cooked P. cocos powder, low-dose and highdose groups of P. cocos surrogate culture powder, low-dose, medium-dose, and high-dose groups of P. cocos water extract, and low-dose, medium-dose, and high-dose groups of P. cocos alcohol extract. Continuous gavage was administered for 7 d. After the final administration, fecal samples from the colon were collected for 16S rDNA sequencing. The results showed that the primary functions of P. cocos extracts obtained through various methods were associated with metabolism, genetic information processing, environmental information processing, cell transformation, and human disease-related genes. Compared with the negative control group, the P. cocos alcohol extract groups exhibited the highest α diversity index, and the high-dose P. cocos alcohol extract group showed the greatest number of operational taxonomic units (OTUs), with Parasutterella (a genus in the phylum Proteobacteria) being the most abundant. In the low-dose raw P. cocos powder group, Akkermansia (a genus in the phylum Verrucomicrobiota) had the highest abundance. Different extraction methods of P.cocos could regulate the intestinal flora of mice and suggested potential improvements in the treatment of obesity and related metabolic diseases.
