中文摘要: 为获得适合蓝莓酒发酵的产香酵母菌株,该文从蓝莓果园土壤、蓝莓果实和叶片中分离纯化,得到33 株纯种酵母,后经嗅闻共获得8 株产香酵母。分别对8 株产香酵母进行香气评价,最终确定1 株产香酵母LM4 为研究对象。经过菌种形态观测和内转录间隔区(internal transcribed spacer,ITS)序列分析,菌株LM4 为葡萄汁有孢汉逊酵母(Hanseniaspora uvarum)。在菌株LM4 的耐受性试验中,该菌株表现出优秀的适应能力,在葡萄糖浓度200~500 g/L、温度18~30 ℃、SO2 浓度50~200 mg/L、pH2.5~4.5 条件下,均能够生长。利用气相色谱质谱联用(gas chromatographymass spectrometry,GC-MS)技术分析该菌株的发酵液挥发性香气成分,检测到17 种挥发性香气成分,香气的主要贡献成分为酯类和醇类化合物。综上,菌株LM4 有望用于蓝莓酒的发酵生产。
Abstract:To obtain an aroma-producing yeast suitable for blueberry wine fermentation,this paper obtained 33 pure yeasts by conducting separation and purification on the soil,fruits,and leaves of blueberry orchards,and then acquired eight aromatic-producing yeasts by olfactory evaluation. Finally,one aroma-producing yeast(LM4) was adopted as the research object. By utilizing observation of the strain morphology and internal transcribed spacer (ITS) sequence analysis,it was determined that LM4 was Hanseniaspora uvarum. In the tolerance experiment of LM4,this strain demonstrated exceptional adaptability and was capable of thriving under glucose concentrations of 200-500 g/L,temperatures of 18-30 ℃,SO2 concentrations of 50-200 mg/L,and pH of 2.5-4.5. Additionally,gas chromatography-mass spectrometry (GC-MS) technology was employed to analyze the volatile aroma components in the fermentation broth produced by this strain,and finally detect 17 volatile aroma components,with the main contributors of ester and alcohol compounds to the aroma. In conclusion,the strain LM4 is expected to be employed in the fermentation production of blueberry wine.
