中文摘要: 该文采用木瓜蛋白酶和中性蛋白酶复合酶法水解汉麻粕获得汉麻多肽,经膜分离技术得到相对分子质量小于1 000 Da 的多肽组分。采用飞行时间质谱仪测定汉麻多肽的相对分子量分布范围。通过构建人肝癌Hep3B 细胞斑马鱼肿瘤细胞模型,研究汉麻多肽体内抗肝癌细胞作用。结果表明,汉麻多肽的相对分子量主要分布在600 Da 以下,能够抑制斑马鱼体内肿瘤细胞生长,抑制肠下新生血管生成,病理组织切片显示汉麻多肽可以明显减少腹腔内肿瘤细胞的数量,改善肿瘤细胞的异常形态。综上,汉麻多肽可作为一种功能活性物质,改善肝癌患者生存品质。
Abstract:This paper employed the papain and neutral protease to hydrolyze hemp meal and thus obtain hemp peptides. Peptides with a relative molecular mass less than 1 000 Da were acquired by membrane separation technology. Meanwhile, the relative molecular weight distribution range of hemp peptides was determined by MALDI-TOF time-of-flight mass spectrometry. The effect of anti-liver cancer cells of hemp peptides in vivo was studied by building a human liver cancer Hep3B cell zebrafish tumor cell model. The results showed that the relative molecular weight of hemp peptides was mainly below 600 Da, thus inhibiting both the growth of tumor cells in zebrafish and the formation of subintestinal neovasculature. Pathological tissue slices revealed that hemp peptides could significantly reduce the number of tumor cells in the abdominal cavity. In conclusion,hemp peptides could be adopted as functional active substances to improve the life quality of patients with liver cancer.
