中文摘要: 醋酸纤维素(cellulose acetate,CA)是一类重要的食品包装材料,但由于其乙酰基含量较高,难以在自然环境中被降解。为提高CA 的降解性能,研究不同酸预处理、氧化预处理和碱预处理对CA 的脱乙酰效果以及单一酶和复配酶对相应脱乙酰样品的酶解效果。结果表明,3 种预处理方法中,NaOH-95%EtOH 碱预处理脱乙酰效果最好。单一酯酶降解不同取代度(degree of substitution,DS)样品后,其失重率呈现相同的规律,即DS1.65>DS1.92>DS2.16。单一纤维素酶降解不同取代度样品196 h 后,DS1.65 的CA 样品总糖含量分别为28.46%(纤维素酶1)、14.35%(纤维素酶2)和22.28%(纤维素酶3),DS1.92 的CA 样品其总糖含量缓慢增加,而DS2.16 的CA 样品总糖含量较低。在复配酶酶解配方中,脂肪酶PS 与纤维素酶1 以及脂肪酶A 与纤维素酶3 复配酶解效果最好,DS1.65 的CA 样品总糖含量比单一纤维素酶酶解分别提高9.64%和15.82%,DS1.92 的CA 样品总糖含量分别提高1.52%和1.89%,而对DS2.16 的CA样品影响不大。综上,复配纤维素酶和脂肪酶能显著提高低取代度样品的降解性。
Abstract:Cellulose acetate (CA), an important material for food packaging, is hard to degrade in natural environments due to its high acetyl content. To enhance the degradability of CA, this study investigated the deacetylation effects of acid pretreatment, oxidation pretreatment, and alkali pretreatment on CA, as well as the enzymatic hydrolysis effects of single enzymes and compound enzymes on the corresponding deacetylated samples. The results showed that the NaOH-95% EtOH alkali pretreatment had the best deacetylation effect among the three pretreatment methods. After the samples with different degrees of substitution (DSs) were degraded by single esterase, the weight loss rate showed the same rule, that is, DS1.65 > DS1.92 > DS2.16. After the samples with different DSs were degraded by single cellulase for 196 h, the total sugar content of the DS1.65 sample was 28.46% (cellulase 1), 14.35% (cellulase 2), and 22.28% (cellulase 3), respectively. The total sugar content of the DS1.92 sample increased slowly, while the total sugar content of the DS1.92 sample was very low. In the formula for compound enzyme hydrolysis, lipase PS with cellulase 1 and lipase A with cellulase 3 had the best hydrolysis effect. Compared with single cellulase hydrolysis, the above two formulas increased the total sugar content of DS1.65 by 9.64% and 15.82%, respectively, increased the total sugar content of DS1.92 by 1.52% and 1.89%, respectively, but had little effect on DS2.16 sample. Therefore, lipase compounded with cellulase can significantly improve the degradability of the samples with low DS.
