中文摘要: 为建立一种脱色评价的方法,以样品吸光度曲线的曲线下面积(area under the curve,AUC)作为脱色率计算依据,将AUC 应用于枸杞多糖的脱色工艺考察中,探究枸杞多糖的最佳脱色工艺条件。采用紫外-可见分光光度计,在200~760 nm 处对3 种色素(原花青素、叶绿素铜钠、β-胡萝卜素)的溶液分别进行全波长扫描,采用Origin 软件统计脱色前后样品液的AUC,用于计算脱色率。将AUC 脱色计算方式引入到枸杞多糖的脱色工艺考察,筛选不同类型大孔树脂的脱色效果,并通过单因素与正交试验确定最佳脱色条件。结果表明:上述3 种色素分别在0.98~31.25、3.92~125.00、0.39~12.50 μg/mL 范围内线性关系良好,R2≥0.998 6,平均加标回收率分别为101.86%、100.08%、100.68%,相对标准偏差(relative standard deviation,RSD)分别为2.66%、2.27%、2.55%。采用AUC 脱色率计算方法,枸杞多糖粗品采用S8 大孔树脂脱色,最佳脱色工艺条件为每100 mg 多糖粗品使用S8 树脂量为7.5 g、粗品浓度为3.0 g/L、在35 ℃下脱色时间为60 min。在该脱色工艺条件下,脱色率的平均值为73.01%,RSD 为1.01%,说明稳定性良好。

Abstract:To establish a new decolorization evaluation method,this paper used the area under the curve(AUC)of the sample absorbance curve as the basis for decolorization rate calculating and applied the AUC to the decolorization process investigation of Lycium barbarum polysaccharide,thus exploring the optimal decolorization process of L. barbarum polysaccharide. The paper performed full wavelength scanning on solutions of three pigments(proanthocyanidins,sodium copper chlorophyll,and beta-carotene)at 200-760 nm through ultraviolet-visible spectrophotometry and calculated the AUC of the sample solutions before and after decolorization through Origin,thus calculating the decolorization rate. The method of AUC decolorization was introduced into the decolorization process investigation of L.barbarum polysaccharide to select the decolorization effects of different types of macroporous resins and determined the optimal decolorization conditions through single factor and orthogonal tests. The results showed that the three pigments had good linear relationships in the ranges of 0.98-31.25,3.92-125.00,and 0.39-12.50 μg/mL respectively,with R2≥0.998 6. The average recovery rates were 101.86%,100.08%,and 100.68% respectively,and the relative standard deviation(RSD)were 2.66%,2.27%,and 2.55% respectively. With the AUC decolorization rate calculating method,the crude L. barbarum polysaccharide was decolorized through S8 macroporous resin. The optimal decolorization process was 7.5 g of S8 resin per 100 mg of crude polysaccharide,with a crude concentration of 3 g/L and a decolorization time of 60 min. Under the optimal decolorization process,the average decolorization rate was 73.01%,and the RSD was 1.01%,indicating good stability.
