中文摘要: 为探究不同提取方式对炒麦芽多糖抗氧化及降血糖活性的影响,分别采用超高压提取(ultrahigh pressure extraction,UHPE)、超声波辅助提取(ultrasonic assisted extraction,UAE)、加热回流提取(heat reflux extraction,HRE)法提取炒麦芽多糖,比较不同提取方式下的多糖得率及结构;通过测定DPPH 自由基、ABTS+自由基、羟自由基清除能力考察其抗氧化活性;采用α-淀粉酶和α-葡萄糖苷酶体外抑制活性试验考察其降血糖活性。结果表明:不同提取方式多糖得率由高到低依次为HRE、UAE、UHPE。3 种方法所获得多糖经过纯化后均不含蛋白质和核酸。HRE 法提取得到的多糖表面呈褶皱条状且没有三螺旋结构;UAE 所得多糖表面凹凸不平,有密集的小条状结构,具有三螺旋结构;UHPE 所得多糖具有多个圆形孔状结构,具有三螺旋结构。红外光谱扫描结果显示,提取方式未改变多糖的特征官能团,但不同提取方式所获得多糖的部分特征峰的吸收发生了蓝移或红移。UAE 提取得到的多糖对DPPH 自由基和ABTS+自由基的清除能力最强,UHPE 次之,HRE 最弱;UHPE 提取得到多糖对羟基自由基的清除能力最强,UAE 次之,HRE 多最弱。HRE 提取得到的多糖对α-淀粉酶和α-葡萄糖苷酶的抑制活性最好,UAE 和UHPE 较弱。
Abstract:To explore the effects of different extraction methods on antioxidant and hypoglycemic activities of polysaccharides from fried malt,the polysaccharides from fried malt were extracted by ultrahigh pressure extraction (UHPE),ultrasonic assisted extraction(UAE),and heat reflux extraction(HRE),respectively. The yield and structure of polysaccharides under different extraction methods were compared. The scavenging ability of polysaccharides on DPPH,ABTS+,and hydroxyl radicals was determined to evaluate their antioxidant activity;the in vitro inhibitory activity test of polysaccharides on α-glucosidase and α-amylase was conducted to study their hypoglycemic effect. The results indicated that the yield of polysaccharides under different extraction methods in descending order was HRE,UAE,and UHPE,and the polysaccharides obtained by the three methods all did not contain protein and nucleic acid after purification. The surface of polysaccharides extracted by the HRE method was wrinkled and striped without a triple helix structure,and polysaccharides extracted by UAE had uneven surfaces and dense small strips with a triple helix structure;the polysaccharides extracted by UHPE had multiple circular pores with a triple helix structure. Fourier transform infrared spectrum showed that extraction methods could not change the characteristic functional groups of polysaccharides,but the absorption of some characteristic peaks of polysaccharides shifted to blue or red. The polysaccharides extracted by UAE had the strongest scavenging capacity for DPPH and ABTS+radicals,followed by the polysaccharides extracted by UHPE and HRE. The polysaccharides extracted by UHPE had the strongest scavenging capacity for hydroxyl radicals,followed by the polysaccharides extracted by UAE and HRE,and the inhibitory activity of polysaccharides extracted by HRE on α-glucosidase and α-amylase was the best,followed by polysaccharides extracted by UAE and UHPE.
