中文摘要: 为提高虾青素(astaxanthin,AST)的水溶性,该研究采用酯酶Est3-14 催化虾青素和戊二酸酐(glutaric anhydride,GA)酯化合成虾青素戊二酸酯(astaxanthin glutarates,AG),包括虾青素戊二酸单酯(astaxanthin glutarate monoester,AGM)和虾青素戊二酸二酯(astaxanthin glutarate diester,AGD),以改善虾青素的水溶性。采用薄层层析色谱、高效液相色谱和质谱对目标产物进行表征,确认虾青素戊二酸酯可经酶催化合成。采用高效液相色谱验证AG 的水溶性,结果表明AG 的水溶性较游离形式虾青素有明显提升。采用单因素试验对催化条件进行优化,确定AG 合成的最优条件为以三氯甲烷为反应溶剂、反应温度50 ℃、AST 与GA 摩尔比1∶4 000、加酶量100 U。在最优条件下反应48 h,虾青素的酯化率达到75.8%,其中,AGM、AGD 的产率分别为29.9%、45.9%。
Abstract:To develop astaxanthin(AST)with increased water solubility,astaxanthin glutarates(AG),including astaxanthin glutarate monoester(AGM)and astaxanthin glutarate diester(AGD)was synthesized via the esterification reaction of AST and glutaric anhydride(GA)catalyzed by Est3-14.Thin-layer chromatography,high performance liquid chromatography,and mass spectrometry were used to characterize AST.The enhanced water solubility of AG compared with that of free AST was proved by high performance liquid chromatography.Additionally,the catalytic conditions were optimized through single factor experiment as follows:chloroform as solvent media,catalytic temperature at 50 ℃,molar ratio of AST to GA at 1∶4 000,and Est3-14 dosage of 100 U.After 48 h of reaction under these conditions,the esterification rate of AST reached 75.8%,and the yield of AGM and AGD were 29.9% and 45.9%,respectively.
