中文摘要: 该研究拟筛选与自淬灭探针互补的寡核苷酸序列,利用双链DNA 的荧光超淬灭机制,构建可视化反应体系,从而实现基于自淬灭探针的环介导等温扩增(loop-mediated isothermal amplification,LAMP)可视化快速检测。结果表明:添加与自淬灭探针完全互补的寡核苷酸序列,体系的背景荧光强度会出现显著下降(P<0.05)。当熔解温度(melting temperature,Tm)值高于室温时,缩短互补序列的长度(从3′端)不会影响其淬灭作用。基于互补序列的荧光超淬灭机制构建的LAMP 可视化反应体系能够有效降低背景荧光,反应结束后表现出易于读取的可视化结果,并成功用于虹鳟鱼的快速检测。综上,合适的互补序列能够降低自淬灭探针的背景荧光,利用该互补序列和自淬灭探针建立低背景荧光的LAMP 体系能够在可视化检测中表现出适用性。
Abstract:This paper aimed to screen an oligonucleotide sequence that was complementary to the self-quenching fluorogenic probe,and construct a visualization system using the fluorescence super-quenching mechanism of double-stranded DNA,to realize the rapid visual detection of loop-mediated isothermal amplification(LAMP)based on the self-quenching fluorogenic probe. The results showed that the background fluorescence intensity of the system was decreased after the addition of an oligonucleotide sequence that was completely complementary to the self-quenching fluorogenic probe(P<0.05). When the melting temperature(Tm)was higher than room temperature,the quenching effect was not impacted by shortening the length of the complementary sequence(from the 3′ end). The LAMP visualization system,based on the fluorescence superquenching mechanism of the complementary sequence,effectively reduced the background fluorescence and provided easy-to-read visualization results after reaction. It has been successfully used for rapid detection of Oncorhynchus mykiss.In summary,an appropriate complementary sequence would reduce the background fluorescence of the self-quenching fluorogenic probe,and it was applicable to establish a low-background fluorescence LAMP system by combining this complementary sequence with the self-quenching fluorogenic probe,which was of certain promotional value.
