中文摘要: 为筛选出适合北苍术固体发酵的菌株,以菌落数和粗多糖得率为筛选指标,获得适合北苍术发酵的乳酸菌,以含水量、接种量和发酵时间为因素,在单因素试验基础上,利用响应面法对发酵工艺进行优化。结果表明,从保存的菌株中共筛选出3 株适合北苍术发酵的乳酸菌,通过生理生化试验以及16S rDNA 测序确定该3 株菌分别为乳酸片球菌(Pediococcus ɑcidilɑctici)和戊糖片球菌(Pediococcus pentosɑceus)。北苍术固态发酵的最佳条件为含水量50%、接种量5.5%、发酵时间2 d。在此条件下,北苍术粗多糖得率达到10.10%,相比未发酵组提高了37.45%。抗氧化试验表明,发酵能够显著提高北苍术粗多糖的DPPH 自由基清除率和羟自由基清除率(P<0.05)。
Abstract:To screen the strains suitable for solid-state fermentation of Atrɑctylodes chinensis,the bacterial count and crude polysaccharide yield were used as screening indicators to obtain lactic acid bacteria for the fermentation of A. chinensis. On the basis of single factor experiments,response surface methodology was adopted to optimize the fermentation process with water content,inoculation amount,and fermentation time as factors.The results showed that three strains were screened and identified as Pediococcus ɑcidilɑctici and Pediococcus pentosɑceus by physiological and biochemical tests and 16S rDNA sequencing.The optimal fermentation conditions were water content 50%,inoculation amount 5.5% and 2 d fermentation. Under these conditions,the crude polysaccharide yield was 10.10%,an increase of 37.45% compared with the unfermented group.The antioxidant test revealed that fermentation significantly increased the scavenging ability of crude polysaccharides from A.chinensis against DPPH and hydroxyl radicals(P<0.05).
