中文摘要: 该研究制备分别特异性识别百菌清、多菌灵的两种单克隆抗体,并建立一种同时快速检测蔬菜中百菌清和多菌灵的胶体金免疫层析法。在最适工作条件下,所建立的方法对百菌清、多菌灵的检测限分别为0.08 ng/mL 和1.95 ng/mL,线性范围分别为0.08~10.00 ng/mL 和1.95~250.00 ng/mL。黄瓜、番茄、包菜3 种样品加标回收率为79.2%~112.1%(百菌清)和63.3%~97.5%(多菌灵),变异系数均低于18.9%,试纸条所需检测时间为20 min,可应用于蔬菜中百菌清、多菌灵残留的现场高通量快速筛查。
Abstract:In this study,two monoclonal antibodies were prepared for the specific recognition of chlorothalonil and carbendazim,and a colloidal gold immunochromatography assay was developed for the rapid simultaneous detection of chlorothalonil and carbendazim in vegetables.Under the optimal conditions,the established method had detection limits of 0.08 ng/mL and 1.95 ng/mL for chlorothalonil and carbendazim,respectively,and the linear ranges were 0.08-10.00 ng/mL and 1.95-250.00 ng/mL,respectively.The recovery rates for cucumber,tomato,and cabbage samples ranged from 79.2% to 112.1%(chlorothalonil)and from 63.3% to 97.5%(carbendazim),with coefficients of variation all below 18.9%. The total detection time of the test strips was 20 min,making it applicable for on-site high-throughput rapid screening of chlorothalonil and carbendazim residues in vegetables.
