中文摘要: 该研究使用水提醇沉法从西瓜皮(Exocarpium Citrulli)中提取3 种不同结构的多糖,并对比它们的体外抗氧化活性,采用DEAE-52 纤维素柱和Sephadex G-200 层析柱对西瓜皮多糖进行分离纯化得到相对分子质量不同的3 种多糖并命名为ECP-1、ECP-2 和ECP-3,使用离子色谱、红外光谱、扫描电子显微镜、刚果红实验、热重(thermal gravimetric analyzer,TGA)等进行分析。结果表明:3 种多糖均为水溶性大分子多糖。ECP-3 的自由基清除能力高于另外两种酸性多糖,这可能与ECP-3 含有较高的糖醛酸有关。
Abstract:The in vitro antioxidant activities of three distinct Exocarpium Citrulli polysaccharides(ECPs)obtained by the water extraction-alcohol precipitation method were compared. Three polysaccharides with different molecular weights,i.e.,ECP-1,ECP-2,and ECP-3,were separated by a DEAE-52 cellulose column and Sephadex G-200,and the relationship between the structure and activity was explored.Ion chromatography,infrared spectroscopy,scanning electron microscopy,Congo red test,and thermogravimetric analysis(TGA)were employed to analyze the polysaccharides.The results demonstrated that all the three macromolecular polysaccharides were water-soluble. ECP-3 had stronger activity for scavenging free radicals than the other two acidic polysaccharides,which may be associated with the fact that ECP-3 contained more uronic acids.
