中文摘要: 通过体外模拟,探究黑果腺肋花楸果提取物胃肠消化过程中的活性成分及抗氧化作用变化规律。采用紫外分光光度法测定黑果腺肋花楸果提取物口腔消化液、胃消化液、肠消化液、肠透析液以及黑果腺肋花楸果提取物溶液5 种样品中总黄酮、多酚、花青素、原花青素含量变化;同时,检测5 种样品对羟基自由基、DPPH 自由基、ABTS+自由基、超氧阴离子自由基的清除能力。结果表明:黑果腺肋花楸果提取物中原花青素含量最高,为(333.86±3.46)mg/g,其次是多酚,花青素含量最少。在模拟消化过程中活性成分变化趋势存在差异,花青素含量在口腔消化液中最多,肠消化液次之,胃消化液和肠透析液中较少;原花青素在口腔消化液中最多,胃消化液中减少,肠消化液和肠透析液中消失。多酚在肠消化液中含量最高,口腔和胃消化液中差距不大,肠透析液中明显减少。模拟消化液清除羟基自由基、DPPH 自由基的变化趋势相似,其中口腔消化液抗氧化能力最强(清除羟基自由基IC50值为71 μL,强于维生素C,清除DPPH 自由基IC50 值为13.7 μL,弱于维生素C),肠消化液抗氧化能力最弱,而清除ABTS+自由基能力的结果略有不同,清除超氧阴离子自由基的能力有待进一步研究。通过研究结果可以看出黑果腺肋花楸果提取物消化产物具有潜在的抗氧化活性,并与相关活性成分含量具有一定相关性。
Abstract:The changes of active components and antioxidant activity of Aronia melanocarpa extract during simulated gastrointestinal digestion in vitro were explored. The content of flavonoids,polyphenols,anthocyanins,and procyanidin in simulated oral,gastric,and intestinal digestive fluids and intestinal dialysate of A.melanocarpa extract and in A. melanocarpa extract was determined by ultraviolet spectrometry. Furthermore,the hydroxyl,DPPH,ABTS+,and superanoxide free radical scavenging activities of these active components were determined. The results showed that procyanidin had the highest content[(333.86±3.46)mg/g]in the A.melanocarpa extract,followed by polyphenols,and anthocyanins had the lowest content.The active components in samples changed differently during the simulated digestion. The content of anthocyanins was the highest in oral digestive fluid,secondary in the intestinal digestive fluid,and least in gastric digestive fluid and intestinal dialysate. Procyanidin was the most abundant in oral digestive fluid,moderate in gastric digestive fluid,and disappeared in the intestinal digestive fluid and intestinal dialysate.Polyphenols had the highest content in intestinal digestive fluids,similar content in oral and gastric digestive fluids,and the lowest content in intestinal dialysate. The simulated digestive fluids scavenged hydroxyl and DPPH radicals in similar trends. The simulated oral digestive fluid had the strongest antioxidant activity,with the IC50 of 71 μL for scavenging hydroxyl radicals,stronger than vitamin C,and the IC50 of 13.7 μL for scavenging DPPH radicals,weaker than vitamin C. The simulated intestinal digestive liquid had the weakest antioxidant ability. However,the digestive fluids showed different ABTS+free radical scavenging activities.Further investigation is needed to study the ability to scavenge superoxide anion radicals.The digestion products of A.melanocarpa extract maintained potential antioxidant activity,which had correlations with the content of active components.
