Total Phenols and Flavonoids in Different Polar Fractions of Plumeria rubra L.Extract and Their Biological Activities

中文摘要: 为研究鸡蛋花95%乙醇提取物的不同极性溶剂萃取物中总酚、总黄酮含量,并对其体外抗菌、抗氧化、抗肿瘤活性进行评价,依次用石油醚、氯仿、乙酸乙酯萃取鸡蛋花醇提取物得到包括水层在内的4 个部位萃取物,测定各部位总酚、总黄酮含量;采用微量肉汤稀释法测定各部位对大肠杆菌、金黄色葡萄球菌、鲍曼不动杆菌、肺炎克雷伯菌、铜绿假单胞菌的最小抑菌浓度(minimum inhibitory concentration,MIC);测试各部位对DPPH·、ABTS+·和·OH 的清除能力;采用细胞计数试剂盒法检测各部位对人肝癌细胞(SMMC?7721,HepG2)、人乳腺癌细胞(MDA?MB?231,MCF?7)、人结肠癌细胞(HCT?116)、人结直肠腺癌上皮细胞(DLD?1)、人恶性黑色素瘤细胞(A375)、人神经胶质瘤细胞(U251)共8 种肿瘤细胞活力的影响。结果表明,各部位中乙酸乙酯萃取物总酚和总黄酮含量最高,分别为(57.73±0.09)mg GAE/g 和(558.88±0.99)mg RE/g,且各部位均有抗菌、抗氧化、抑制肿瘤细胞活力的能力,且均呈明显量效关系。其中,乙酸乙酯部位的抗菌、抗氧化能力最强,对5 种细菌的MIC 为0.5~1.0 mg/mL,清除DPPH·、ABTS+·和·OH 的IC50 值分别为14.94、15.43、74.61 μg/mL;氯仿部位对8 种肿瘤细胞的抑制能力最强,其IC50 值为9.12~40.14μg/mL。综上,鸡蛋花各部位均具有较强的抗菌、抗氧化以及抑制肿瘤细胞活性的作用,为鸡蛋花开发利用成食品、保健品或者药品提供参考。

Abstract:This paper aimed to study the total phenols and flavonoids in different polar solvents of the 95%ethanol extract of Plumeria rubra L.,and evaluate their antibacterial,antioxidant and antitumor activities in vi?tro.Petroleum ether,chloroform,ethyl acetate and water were used to extract four active substances from the ethanol extracts of P.rubra and the contents of total phenols and total flavonoids in each fraction were deter?mined.The minimum inhibitory concentration(MIC)of each fraction against Escherichia coli,Staphylococcus aureus,Acinetobacter baumannii,Klebsiella pneumoniae and Pseudomonas aeruginosa was determined by the broth dilution method,and the scavenging capacity against DPPH·,ABTS+·and·OH was measured.In addi?tion,the anti?proliferation effect of each fraction on human hepatocellular carcinoma cells(SMMC?7721,HepG2),human breast cancer cells(MDA?MB?231,MCF?7),human colon cancer cells(HCT?116),hu?man colorectal adenocarcinoma epithelial cells(DLD?1),human malignant melanoma cells(A375)and hu?man glioma cells(U251)was evaluated by Cell Counting Kit?8 assay.The results showed that the highest con?tents of total phenols and total flavonoids were obtained from ethyl acetate fraction,which were(57.73±0.09)mg GAE/g and(558.88±0.99)mg RE/g,respectively.All P.rubra extracts had antioxidant,antibacterial and antitumor abilities and was dose?dependent with the concentration.Among them,the ethyl acetate fraction had the strongest antibacterial and antioxidant abilities,with the MIC against the five bacteria ranging from 0.5 mg/mL to 1.0 mg/mL and the IC50 of scavenging DPPH·,ABTS+· and ·OH being 14.94,15.43 and 74.61 μg/mL,respectively.The chloroform fraction inhibited the proliferation of the above eight tumor cells most,with the IC50 ranging from 9.12 μg/mL to 40.14 μg/mL.In conclusion,each fraction of P.rubra has strong antibacterial,antioxidant and antitumor activities,which provides a certain theoretical basis for the de?velopment and utilization of P.rubra as food health care products or drugs.
