中文摘要: MYB 转录因子广泛存在于真核生物中,在植物的生长发育、逆境胁迫等过程中发挥重要作用。与植物相比,对食用菌中MYB 转录因子的研究有限。为探究MYB 转录因子在食用菌中的生物学功能,对前期转录组发现的一个编码双孢蘑菇(Agaricus bisporus)MYB 转录因子的基因进行克隆和生物信息学分析。结果表明,该基因编码区长1 209 bp,翻译402 个氨基酸;编码的蛋白分子量为45.95 kDa,等电点9.17,是一种不存在跨膜结构、不含信号肽的亲水性蛋白,具有4 个高度保守的SANT 结构域,属于4R 型MYB 转录因子,与白环蘑(Leucoagaricus)中的4RMYB 转录因子亲缘关系最近;二级结构预测显示以无规则卷曲和α-螺旋结构为主;亚细胞定位分析显示该转录因子位于细胞核中;此外启动子序列分析表明,该基因启动子区域含有多个与生物抗逆应答、激素响应等相关的顺式作用元件。
Abstract:MYB transcription factors are ubiquitous in eukaryotes and play a key role in plant growth,development,and stress responses.Although extensive studies have been carried out for the MYB transcription factors in plants,little is known about the MYB transcription factors in edible mushrooms.A MYB transcription factor gene in Agaricus bisporus that was identified in the previous transcriptome study was cloned and bioinformatically analyzed.The results showed that the gene had a coding region of 1 209 bp,which encoded 402 amino acid residues.The deduced protein had a molecular weight of 45.95 kDa and an isoelectric point of 9.17,being a hydrophilic protein with no transmembrane structure or signal peptide.In addition,the protein had four highly conserved SANT domains,being a 4R-MYB transcription factor.It had the closest relationship with the MYB transcription factor in Leucoagaricus.The secondary structural elements in the deduced protein were mainly random coils and α-helices.The subcellular localization analysis showed that the protein was located in the nucleus.Furthermore,the promoter region of this gene contained multiple cis-acting elements associated with biotic stress responses and hormone responses.
