中文摘要: 采用抗性淀粉癸酸复合物(resistant starch-capric acid,RS-CA)实现癸酸在结肠的定点释放,研究癸酸对脂多糖(lipopolysaccharide,LPS)引起的小鼠抑郁症的改善作用。选取40 只雄性ICR 小鼠随机分成5 组(空白组、LPS组、LPS+RS 组、LPS+氟西汀组、LPS+RS-CA 组)。所有干预样品均以体积比2∶1 溶解在小鼠的饮水中。最后一次LPS 腹腔注射后,进行行为学实验;随后处死动物,眼球取血保留海马,用于生化指标检测;保留结肠和脑组织进行苏木精-伊红染色;收集粪便进行16S rRNA 检测。结果表明,经RS-CA 预处理的小鼠在悬尾实验和旷场实验中表现良好,同时降低了抑郁小鼠血清肿瘤坏死因子-α(tumor necrosis factor-α,TNF-α)水平,提高了海马5-羟色胺(5-hydroxytryptamine,5-HT)水平,表明RS-CA 对LPS 造成的结肠腺细胞杂乱排列以及海马神经元核浓缩和嗜酸变性具有改善作用。16S rRNA 结果表明,RS-CA 预处理逆转了与抑郁症相关菌群的失衡,包括Firmicutes/Bacteroidetes 比值降低,Prevotella、Parabacteroides、Klebsiella 和Acfinobacteria 恢复至正常水平;同时改善了肠道菌群的均匀性,使肠道微生物组成与正常组一致。以上结果表明,RS-CA 可以通过调节肠道菌群降低下丘脑-垂体-肾上腺素(hypothalamic-pituitary-adrenal axis,HPA)轴炎症反应从而实现抗抑郁作用。

Abstract:This paper achieved the fixed point release of capric acid in the colon using resistant starch-capric acid(RS-CA),to study the improvement effect of capric acid on lipopolysaccharide(LPS)-induced depression in mice. Forty male ICR mice were randomly divided into five groups(blank group,LPS group,LPS+RS group,LPS+fluoxetine group,and LPS+RS -CA group). All the intervention samples were dissolved in the drinking water of mice with volume ratio of 2∶1. Behavioral test was performed after the last intraperitoneal injection of LPS. Then the mice were sacrificed,and the eyeball blood was collected to retain the hippocampus for biochemical index detection. The colon and brain tissues were retained for HE staining. Feces were collected for 16S rRNA detection. The results showed that the mice pretreated with RS-CA performed well in the tail suspension test and open field test,and had reduced serum tumor necrosis factor- α(TNF-α)and increased hippocampus 5-hydroxytryptamine(5-HT),this indicates that RS-CA could improve the disordered arrangement of colon adenocarcinoma cells as well as the nucleus concentration and acidophilic degeneration of hippocampal neurons caused by LPS. The 16S rRNA detection revealed that RS-CA pretreatment reversed the imbalance of bacteria associated with depression;for example,Firmicutes/Bacteroidetes decreased and Prevotella,Parabacteroides,Klebsiella and Acfinobacteria returned to normal level. Additionally,the homogeneity of intestinal microbiota was improved,and thus mice in LPS+RS-CA group had the same intestinal microflora with those in blank group. The above results indicated that RS-CA could achieve antidepressant effect by regulating intestinal microbiota and reducing the inflammatory response of the hypothalamic-pituitary-adrenal axis(HPA).
