中文摘要: 为促进核桃分心木多酚(Diaphragma juglandis Fructus polyphenols,DJF-P)的开发,该文通过对比5 种大孔树脂对DJF-P 静态吸附解吸效果,筛选出AB-8 型大孔树脂最适于纯化DJF-P 溶液,同时优化大孔树脂纯化的单因素试验条件,并通过高效液相色谱法(high performance liquid chromatography,HPLC)进一步对DJF-P 溶液纯化前后的物质组成成分进行分析。结果表明:最佳提取条件为55%乙醇,料液比为1∶15(g/mL),室温下浸提24 h,浓缩,得到DJFP 浓缩液。最佳纯化工艺为55%乙醇稀释DJF-P 浓缩液20 倍,AB-8 型大孔树脂静态吸附解吸时间均为8 h,解吸液为70%乙醇。HPLC 结果表明,DJF-P 含有槲皮素、阿魏酸、芦丁、绿原酸和没食子酸。

Abstract:In order to promote the development of Diaphragma juglandis Fructus polyphenols(DJF-P),This study adopted a comparative analysis of five types of macroporous resins regarding their static adsorption-desorption performance for DJF-P.AB-8 macroporous resin was selected as the most suitable resin for purification of DJF-P solutions,and the single-factor experimental conditions for macroporous resin purification were optimized.The compositional components of DJF-P solutions before and after purification was further analyzed by high performance liquid chromatography(HPLC).The results indicated that the optimal extraction conditions were 55% ethanol at a solid-liquid ratio of 1∶15(g/mL),extraction at room temperature for 24 h,and subsequent concentration to obtain DJF-P concentrate.The optimal purification process included diluting the concentrated DJF-P solution with 55% ethanol at a 20 fold dilution,with an 8 h of static adsorption-desorption time using AB-8 macroporous resin,and a desorption solution comprising 70% ethanol.HPLC results showed that DJF-P contained quercetin,ferulic acid,rutin,chlorogenic acid,and gallic acid.
