中文摘要: 为深入探究小米谷糠多糖的抗氧化性和免疫活性,该研究采用水提醇沉法制备小米粗多糖(millet polysaccharide,MP),经DEAE-52 纤维素层析分离纯化得到组分MP-1 和MP-2,并对分离前后多糖的化学成分、分子量、抗氧化能力及免疫活性进行评估。结果表明:小米谷糠MP、MP-1 和MP-2 的多糖含量分别为59.78%、78.58%和45.78%,分子量分别为16.4、14.1、13.9 kDa;体外抗氧化实验表明小米谷糠多糖具有显著的自由基清除能力;细胞实验表明MP-1 具有更显著的免疫调节能力,能够促进NO 产生及RAW264.7 细胞增殖,进而具有免疫调节活性;逆转录聚合酶链式反应(reverse transcription-polymerase chain reaction,RT-PCR)结果表明小米谷糠多糖通过与TLR4 受体特异性结合进而促进巨噬细胞的活化,并促进炎症因子的分泌。小米谷糠多糖MP-1 不仅可以作为抗氧化剂,更可以作为一种天然来源的功能性食品营养补充剂。
Abstract:In this study,in order to further explore the antioxidant and immune activities of millet bran polysaccharide,millet polysaccharide(MP)was prepared by water extraction and alcohol precipitation method.MP-1 and MP-2 were separated and purified by DEAE -52 cellulose chromatography.The chemical composition,molecular weight,antioxidant capacity and immune activity of MP before and after separation were evaluated.The results showed that the polysaccharide content of MP,MP-1 and MP-2 was 59.78%,78.58% and 45.78%,respectively.The molecular weights were 16.4,14.1 kDa and 13.9 kDa.In vitro antioxidant experiments showed that millet bran polysaccharide had significant antioxidant activity and free radical scavenging ability.Cell experiments showed that MP-1 had more significant immunomodulatory ability,and it could promote the production of NO and the proliferation of RAW264.7 cells,thus showing immunomodulatory activity.Reverse transcription-polymerase chain reaction(RT-PCR)results indicated that millet bran polysaccharide could promote the activation of macrophages and secretion of inflammatory factors by specifically binding to TLR4 receptor.Millet bran polysaccharide MP-1 could not only be used as an antioxidant,but also as a potential functional dietary/nutritional supplement and natural source.
