中文摘要: 该研究选用头孢哌酮诱导的艰难梭菌肠炎模型,探究岩藻聚糖对抗生素相关的艰难梭菌结肠炎的预防活性。结果表明,预先服用头孢哌酮期间同时口服300 mg/kg 岩藻聚糖(重均分子量126.65 kDa)10 d,可有效缓解艰难梭菌侵染引起的腹泻和结肠黏膜损伤。盲肠的菌群结构分析发现,口服岩藻聚糖能够明显抑制盲肠中肠炎相关的艰难梭菌和肠球菌的相对丰度,提高活泼瘤胃球菌、布劳特氏菌属等有益菌的相对丰度,同时减少盲肠中艰难梭菌毒素A(Clostridium difficile toxin A,Tcd A)和艰难梭菌毒素B(Clostridium difficile toxin B,Tcd B)的产生,但是对肠道菌群的丰度和多样性的恢复作用很小。岩藻聚糖还能减少结肠中巨噬细胞和中性粒细胞的浸润,下调促炎因子肿瘤坏死因子-α(tumour necrosis factor alpha,TNF-α)、干扰素-γ(interferon gama,IFN-γ)、白介素-1β(interleukin 1 beta,IL-1β)和白介素-6(interleukin 6,IL-6)的基因表达,上调抗炎因子白介素-4(interleukin 4,IL-4)和白介素-10(interleukin 10,IL-10)的基因表达。综上,岩藻聚糖不能恢复肠道菌群的丰度和结构,其预防艰难梭菌肠炎活性与抑制艰难梭菌的侵染、产毒活性、抗炎活性以及肠黏膜保护活性有关。因此,岩藻聚糖可以用于预防头孢哌酮相关的艰难梭菌肠炎。
Abstract:In this study,the model of Clostridioides difficile-associated colitis(C.difficile colitis)induced by cefoperazone was used to explore the preventive activity of fucoidan against C.difficile colitis related to antibiotics.The result showed that oral administration of 300 mg/kg fucoidan(weight-average molecular weight 126.65 kDa)for 10 d with simultaneous cefoperazone treatment could effectively alleviate diarrhea and colonic mucosal damage caused by C.difficile.The analysis of the gut microbiota community demonstrated that taking fucoidan orally could effectively inhibit the relative abundance of C.difficile and Enterococcus associated with colitis,increase the relative abundance of beneficial bacteria,such as Ruminococcus gauvreauii group and Blautia in the cecum,and reduce C.difficile toxin A (Tcd A)and B (Tcd B).However,fucoidan treatment only had a limited effect on restoring the abundance and diversity of gut microbiota.Fucoidan could also reduce the infiltration of macrophages and neutrophils in the colon,down-regulate the gene expressions of proinflammatory factors,including tumour necrosis factor alpha(TNF-α),interferon gama(IFN-γ),interleukin 1 beta(IL-1β),and interleukin 6(IL-6),and up-regulate those of anti-inflammatory factors,namely interleukin 4 (IL-4)and interleukin 10 (IL-10).In conclusion,fucoidan failed to restore the abundance and structure of gut microbiota,and its preventive activity against C.difficile colitis was related to inhibition on C.difficile infection or toxin production,anti-inflammation,and colonic mucosal protection.Therefore,fucoidan can be used for preventing C.difficile colitis induced by cefoperazone.
