中文摘要: 为探究不同剂量表没食子儿茶素没食子酸酯(epigallocatechin gallate,EGCG)对高脂饮食诱导的代谢综合征小鼠的干预作用,将小鼠分为正常饮食对照组、高脂饮食对照组、低剂量EGCG 干预组、中剂量EGCG 干预组及高剂量EGCG 干预组,其中低、中、高EGCG 灌胃剂量分别为50、100、200 mg/kg b.w.,每周监测体质量,连续灌胃12 周后测定小鼠的血清生理生化指标、检测回肠组织中脂质代谢相关基因的表达量以及肠道氧化应激指标。结果表明,不同剂量EGCG 干预均可显著降低高脂饮食小鼠体质量的增加并改善脂质代谢异常,这可能与降低肠道中脂质吸收相关基因的表达有关,但中、高剂量EGCG 可能具有一定的肠毒性。
Abstract:The intervention effects of different doses of epigallocatechin gallate(EGCG)on high-fat-diet-induced metabolic syndrome in mice were studied. The mice were assigned into a control(a normal diet)group,a model(a high-fat diet)group,and low-,medium-,and high-dose(50,100 mg/kg b.w. and 200 mg/kg b.w.,respectively)EGCG groups. The body weights of mice were measured weekly. After 12 weeks of gavage,the mice were euthanized. Tissues and serum samples were collected for the determination of biochemical indicators,mRNA levels of genes involved in lipid metabolism in the ileum,and oxidative stress-associated indicators. The results showed that different doses of EGCG significantly reduced the weight gain and ameliorated dyslipidemia in the mice fed with a high-fat diet,which might be attributed to the down-regulated mRNA levels pf the genes involved in lipid absorption in the ileum. However,medium and high doses of EGCG might induce intestinal toxicity.
