中文摘要: 非特异性过氧合酶(unspecific peroxygenase,UPO)能够利用过氧化氢(H2O2)催化多种氧化反应,在工业中具有很大的应用潜力。针对AaeUPO 在毕赤酵母中表达量低以及发酵工艺不清的问题,通过摇瓶发酵优化以及正交试验确定发酵培养基条件,并进一步通过5 L 发酵罐进行放大。结果表明,毕赤酵母摇瓶水平高效表达AaeUPO 的基本发酵条件:甲醇浓度1.00%、培养基初始pH5.5、蛋白胨浓度1.5%、酵母粉浓度1.5%,最优发酵条件下粗酶液酶活达到18.2 U/mL。在摇瓶水平发酵试验的基础上进行5 L 发酵罐放大试验,优化后以接种量10%、诱导温度30 ℃、流加甲醇作为最终工艺条件。最终得到的细胞湿重244 g/L,最高酶活为25.1 U/mL,相较于初期未优化酶活提升了91.6%。将AaeUPO 用于H2O2 检测,AaeUPO 对于H2O2 具有较好的检测性能,检测限达到5 μmol/L,抗干扰性能强。
Abstract:Unspecific peroxygenase(UPO)was capable of catalyzing various oxidation reactions using hydrogen peroxide(H2O2)and had great potential for industrial applications.In order to address the low expression level of AaeUPO in Pichia pastoris and the unclear fermentation process,the fermentation conditions were optimized through shake flask fermentation and orthogonal tests,and further scaled up using a 5 L fermenter.The results showed that the basic fermentation conditions for high-efficiency expression of AaeUPO in P.pastoris shake flask fermentation were as follows:methanol concentration of 1.00%,initial pH value of 5.5,peptone concentration of 1.5%,and yeast powder concentration of 1.5%.Under the optimal fermentation conditions,the crude enzyme activity reached 18.2 U/mL.Based on the shake flask fermentation experiment,a 5 L fermenter was used for scale up,and the final process conditions were optimized with an inoculation volume of 10% ,induction temperature of 30 ℃,and methanol fed-batch addition.The resulting cell wet weight was 244 g/L,and the highest enzyme activity reached 25.1 U/mL,which was a 91.6% improvement compared with the non-optimized initial stage.AaeUPO was used for H2O2 detection and demonstrated good detection performance with a detection limit of 5 μmol/L and strong anti-interference capability.
