中文摘要: 为研究茶碱和可可碱对高脂饮食诱导小鼠体质量和胰岛素抵抗的影响,选取50 只健康的C57BL/6J 小鼠,随机分为低脂饮食(low-fat diet,LFD)组、LFD+茶碱组、LFD+可可碱组、高脂饮食(high-fat diet,HFD)组、HFD+茶碱组、HFD+可可碱组。每周定期检测小鼠体质量、空腹血糖水平、摄食量和饮水量,14 周和15 周进行腹腔葡萄糖耐量试验(intraperitoneal glucose tolerance test,IPGTT)和腹腔胰岛素耐受试验(intraperitoneal insulin tolerance test,IPITT),饲养结束后测定脏器系数和血清生化指标。结果显示:茶碱能有效降低小鼠体质量(低于LFD 组4.89%~16.28%,HFD 组7.86%~30.81%)和减少脂肪堆积;IPGTT 和IPITT 表明茶碱能改善小鼠葡萄糖耐受性和缓解胰岛素抵抗;与HFD 组相比,茶碱能降低小鼠血清总胆固醇、甘油三酯、低密度脂蛋白和谷丙转氨酶水平,但可可碱没有发挥这些作用。综上,茶碱和可可碱虽互为同分异构体的甲基黄嘌呤生物碱,但对HFD 诱导小鼠在减肥降脂方面的影响相差较大。其中,茶碱能抑制HFD 诱导小鼠的体质量增加和缓解胰岛素抵抗,而可可碱没有产生此类效果。
Abstract:The effects of theophylline and theobromine on the body weight and insulin resistance of the mice fed with a high-fat diet(HFD)were studied.Fifty healthy C57BL/6J mice were selected and randomly divided into low-fat diet(LFD)group,LFD+theophylline group,LFD+theobromine group,high-fat diet(HFD)group,HFD+theophylline group,and HFD+theobromine group.The body mass,fasting blood glucose level,food intake,and water intake were recorded every week.The intraperitoneal glucose tolerance test(IPGTT)and intraperitoneal insulin tolerance test(IPITT)were performed at weeks 14 and 15.Organ indexes and serum biochemical indexes were determined at the end of the feeding treatment.The results showed that theophylline reduced the body mass by 4.89%-16.28% compared with the LFD group and by 7.86%-30.81% compared with the HFD group and inhibited fat accumulation.The results of IPGTT and IPITT showed that theophylline improved glucose tolerance and alleviated insulin resistance in mice.Compared with the HFD group,theophylline lowered the serum levels of total cholesterol,triglycerides,low-density lipoprotein,and alanine aminotransferase in mice,while theobromine did not exert these effects.In conclusion,although theophylline and theobromine,as methylxanthine alkaloids,are isomers of each other,their effects on weight loss and lipid reduction in HFD-induced mice are different.Theophylline can reduce the body mass gain and alleviate insulin resistance in HFD-induced mice,while theobromine did not show such effects.
