苦丁茶结合酚的提取优化及其对α-葡萄糖苷酶的抑制作用

中文摘要: 以苦丁茶为原料,根据单因素试验结果,通过响应面法优化苦丁茶结合酚的最佳提取条件,采用不同型号大孔树脂对苦丁茶结合酚进行吸附,以除去粗提物中的盐,并且研究其对淀粉消化酶α-葡萄糖苷酶的抑制作用。试验表明,苦丁茶结合酚最佳提取条件为氢氧化钠浓度8.5 mol/L、液料比43∶1(mL/g)、提取时间4.2 h,此优化条件下,测得结合酚含量为(29.8±2.0)mg/g;7 种不同类型的大孔树脂中,AB-8 大孔树脂对苦丁茶结合酚的吸附率和解吸率最高;苦丁茶结合酚对α-葡萄糖苷酶的抑制作用明显,其半抑制浓度为1.90 mg/mL,抑制类型为可逆性抑制,并且是非竞争性抑制类型,抑制常数Ki 为0.60 mg/mL。
Abstract:The optimal extraction conditions for Kuding tea(KT)binding polyphenols were selected using response surface methodology based on the results of single-factor experiment.Different types of macroporous resins were used to adsorb KT binding polyphenols to remove salts from crude extracts,and their inhibitory effects on the digestion enzyme α-glucosidase were studied.The results showed that the optimal extraction conditions for KT binding polyphenols were as follows:sodium hydroxide concentration of 8.5 mol/L,liquid-to-material ratio of 43∶1(mL/g),and extraction time of 4.2 h.Under these optimized conditions,the content of polyphenols was determined to be(29.8±2.0)mg/g.Among the seven different types of macroporous resins tested,AB-8 macroporous resin exhibited the highest adsorption rate and desorption rate for KT binding polyphenols.KT binding polyphenols showed significant inhibition on α-glucosidase,with a half-inhibitory concentration of 1.90 mg/mL,reversible inhibition,and non-competitive inhibition with an inhibition constant Ki of 0.60 mg/mL.
