中文摘要: 为探究滇皂角米多糖碱提工艺,以多糖提取率为指标,通过单因素试验考察液料比、NaOH 浓度、提取温度及提取时间对滇皂角米多糖提取率的影响,结合Box-Behnken 响应面试验对提取工艺进行优化,并对多糖进行体外抗氧化探究。结果表明,滇皂角米多糖的碱提最佳工艺:液料比220∶1(mL/g)、NaOH 溶液浓度0.16 mol/L、提取温度51 ℃、提取时间90 min,在该条件下滇皂角米多糖的提取率为(73.64±0.56)%。此外,滇皂角米多糖具有一定的体外抗氧化能力,清除DPPH 自由基、ABTS+自由基的IC50 值分别为10.87、6.67 mg/mL。
Abstract:To investigate the alkalinity extraction technology of polysaccharides from the seeds of Gleditsia japonica,the extraction yields were used as indicators for the effects of four factors.These included the liquidsolid ratio NaOH solution concentration,extraction temperature,and extraction time on the efficacy of polysaccharides from the seeds of Gleditsia japonica,using single-factor tests.Based on the experimental results,Box-Behnken response surface methods were conducted to optimize the extraction process.In addition,the antioxidant capacities (in vitro)of the extracts were analyzed.The results showed that the optimum extraction conditions included:a liquid-solid ratio of 220∶1 (mL/g),alkali concentration of 0.16 mol/L,extraction temperature of 51 ℃,and extraction time of 90 min.Under these conditions,the extraction yield was(73.64±0.56)%.Moreover,the polysaccharides possessed certain antioxidant capacities in vitro.The IC50 values for scavenging DPPH and ABTS+free radicals were 10.87 mg/mL and 6.67 mg/mL,respectively.
