中文摘要: 采用超声辅助提取法开展多花黄精多糖提取工艺研究,在单因素试验的基础上,以多糖得率为指标,响应面法优化料液比、超声功率、超声时间、超声温度提取工艺参数;采用响应面设计优化多花黄精多糖含片配方,以综合评分为指标,在考察单因素基础上,获得最佳的填充剂、矫味剂、润滑剂添加量以及制粒润湿剂的体积分数。结果表明,多花黄精多糖超声辅助提取法最佳提取工艺参数为料液比为1∶40(g/mL)、超声功率为270 W、超声温度为71 ℃、超声时间为49 min,该条件下多花黄精多糖提取的得率为(25.07±0.15)%。多花黄精多糖含片最佳配方为填充剂添加量19.91%(甘露醇:微晶纤维素=3∶1,质量比)、矫味剂木糖醇添加量14.55%、矫味剂柠檬酸添加量5.04%、润滑剂硬脂酸镁添加量0.5%、制粒润湿剂乙醇的体积分数为85%。多花黄精多糖提取及其含片制备工艺稳定可控,多花黄精多糖含片中黄精多糖含量不低于0.28 g/片,崩解时限、脆碎度符合质量要求,呈淡黄色,色泽均匀,酸甜适宜。
Abstract:The ultrasonic assisted extraction method was adopted to study the extraction process of Polygonatum cyrtonema Hua polysaccharides.On the basis of single-factor experiment and with the yield of polysaccharides as the index,the extraction parameters of solid-liquid ratio,ultrasonic power,ultrasonic time and ultrasonic temperature were optimized by response surface method;The formulation of polysaccharides buccal tablets of P.cyrtonema Hua was optimized by response surface design,the comprehensive score was used as the index to obtain the optimal addictive amount of fillers,flavor enhancers and lubricants as well as the volume fraction of moistening agents for granulation on the basis of single-factor experiment.The results showed that the optimum ultrasonic assisted extraction method parameters were as follows:the solid-liquid ratio of 1∶40(g/mL),ultrasonic power of 270 W,ultrasonic temperature of 71℃and ultrasonic time of 49 min.Under this condition,the yield of P.cyrtonema Hua polysaccharides was(25.07±0.15)%.The optimal formulation of polysaccharides buccal tablets of P. cyrtonema Hua was as follows:19.91% fillers (mannitol:microcrystalline cellulose=3 ∶1,mass/ratio),14.55% xylitol(flavor enhancer),5.04% citric acid(flavor enhancer),and 0.5% magnesium stearate(lubricant)were added,and the volume fraction of ethanol(moistening agent)was 85%.The extraction of P.cyrtonema Hua polysaccharides and the preparation process of its buccal tablets are stable and controllable. The content of polygonum polysaccharides in the polysaccharides buccal tablets of P. cyrtonema Hua is not less than 0.28 g/tablet,and the disintegration time and fragility meet the quality requirements.The tablets had uniform light yellow color as well as suitable sweet and sour taste.
