树莓果肉多糖提取工艺优化及其抗疲劳作用

中文摘要: 为评价树莓果肉多糖的疲劳活性,通过响应面法优化纤维素酶酶解提取树莓果肉多糖工艺。结果表明,通过Plackett-Burman主因素筛选实验,得到影响树莓果肉多糖得率的3个主效应因素:酶解温度、酶解时间和加酶量。利用响应面法优化得到树莓果肉多糖的最佳工艺为料液比1∶20(g/mL)、pH5、加酶量0.85% 、酶解温度44℃、酶解时间1.4 h。在最佳工艺条件下,得到纤维素酶酶解树莓果肉多糖得率为2.73% 。通过建立小鼠负重游泳实验模型,纤维素酶酶解得到的树莓果肉多糖能够显著延长小鼠负重游泳时间,提高机体的运动耐力,并使小鼠体内的肝糖原及肌糖原水平显著提升,同时降低小鼠血清中尿素氮、乳酸含量,具有较好的抗疲劳效果。
Abstract:The enzymatic hydrolysis process of raspberry pulp polysaccharide was optimized by response surface methodology,and then the anti-fatigue activity of the polysaccharide was studied.Plackett-Burman design was employed to screen out the main factors affecting the yield of raspberry polysaccharide,which were enzymatic hydrolysis temperature,enzymatic hydrolysis time,and enzyme dosage.The enzymatic hydrolysis conditions of polysaccharides from raspberry were optimized by response surface methodology.The optimum conditions were material-to-liquid ratio of 1∶20(g/mL),pH5,enzyme dosage of 0.85% ,and enzymatic hydrolysis at 44℃for 1.4 h.Under the optimal conditions,the yield of polysaccharides extracted from raspberry by hydrolysis with cellulase was 2.73% .The results of weight-bearing test with mice showed that the raspberry pulp polysaccharide obtained by hydrolysis with cellulase prolonged the time of weight-bearing swimming,improved the exercise endurance,significantly elevated the levels of liver glycogen and muscle glycogen,and lowered the serum levels of urea nitrogen and lactic acid in mice.The raspberry pulp polysaccharide had good anti-fatigue effect.
