中文摘要: 该文建立高效液相色谱(high performance liquid chromatography,HPLC)法测定饮料中甜菊醇的定量分析方法,并优化饮料中甜菊糖苷水解成甜菊醇的条件。采用3 mmol/g路易斯酸水解,无水乙醇提取,以70%乙腈水溶液(体积分数)作为流动相,使用ZORBAX SB-C18色谱柱。结果表明,甜菊醇在5.0 μg/mL~100.0 μg/mL的浓度范围内线性关系较好(r2≥0.999),平均回收率在81.9%~92.3%,相对标准偏差在1.17%~3.67%。饮料中甜菊糖苷转化甜菊醇的最佳工艺条件:催化剂为氯化铁,提取温度100℃,甜菊糖苷浓度300 mg/mL,提取时间3 h,催化剂含量3 mmol/g。
Abstract:A high performance liquid chromatography (HPLC)method was developed for the determination of steviol in beverage,and the conditions for the hydrolysis of stevioside into steviol were optimized.Specifically,the samples were hydrolyzed by 3 mmol/g Lewis acid and then extracted by 100%ethanol.HPLC was performed with ZORBAX SB-C18 column and 70%(volume fraction)acetonitrile aqueous solution as the mobile phase.The result suggested that the steviol had good linear relationship (r2≥0.999)within the concentration range of 5.0 μg/mL-100.0 μg/mL.The average recovery ranged from 81.9%to 92.3%,and the RSD from 1.17%to 3.67%.The hydrolysis conditions were optimized as hydrolysis with 3 mmol/g ferric chloride as the catalyst and the stevioside concentration of 300 mg/mL at 100℃for 3 h.
