中文摘要: 以蛹虫草子实体为原料,采用响应面设计优化蛹虫草多酚提取工艺,分析蛹虫草多酚的抗氧化活性。以多酚的得率为指标,在单因素试验基础上,利用Box-Behnken设计进行响应面试验,确定蛹虫草多酚最佳提取工艺,并对多酚体外总抗氧化能力及DPPH自由基、羟自由基清除能力进行分析。结果显示:蛹虫草多酚最佳提取工艺为提取温度51℃、提取时间1.6 h、液料比49∶1(mL/g),优化条件下多酚得率为6.03 mg/g。抗氧化活性分析结果表明:浓度为0~0.4 mg/mL时,蛹虫草多酚总抗氧化能力随着质量浓度的增加而增强,且始终高于同等浓度的维生素C溶液;多酚溶液对DPPH自由基的半抑制质量浓度为19.52 μg/mL,为维生素C溶液的75.25%;对羟自由基半抑制质量浓度为86.47 μg/mL,是维生素C溶液的8.31%。蛹虫草多酚的提取工艺可行,蛹虫草多酚具有较强的抗氧化活性。

Abstract:Using Cordyceps militaris fruiting body as raw material,this paper used response surface methodology(RSM)to optimize the extraction process of C.militaris polyphenols and analyzed the antioxidant activity of C.militaris polyphenols.Based on single factor test,using yield of polyphenols as index,this paper used Box-Behnken design to conduct RSM and determine the optimal extraction process of C.militaris polyphenols.The total antioxidant capacity,DPPH free radical scavenging ability,and hydroxyl free radical scavenging ability of polyphenols in vitro were analyzed.The results showed that the optimal extraction process of C.militaris polyphenols was 51 ℃ of extraction temperature,1.6 h of extraction time,and 49∶1(mL/g)of liquid-solid ratio,and the yield of polyphenols was 6.03 mg/g under optimized conditions.The results of antioxidant activity analysis showed that the total antioxidant capacity of C.militaris polyphenols increased with the increase in mass concentration in the range of 0-0.4 mg/mL,and was always higher than that of vitamin C solution in the same concentration.The semi-inhibitory mass concentration of DPPH free radical in polyphenol solution was 19.52 μg/mL,which was 75.25%of that in vitamin C solution,and the semi-inhibitory mass concentration of hydroxyl radical was 86.47μg/mL,which was 8.31%of that in vitamin C solution.The extraction process of C.militaris polyphenols is feasible,and C.militaris polyphenols has relatively strong antioxidant activity.
