中文摘要: 为探究南瓜籽蛋白肽对D-半乳糖诱导的衰老模型小鼠的抗衰老作用,该研究从不同分子量南瓜籽蛋白肽中筛选出抗氧化活性最强的组分进行动物实验。实验设正常对照组,模型对照组,南瓜籽蛋白肽低、中、高剂量[250、500、1 000 mg/(kg·d)]组。灌胃42 d后测定小鼠的体质量、脏器系数、代谢产物(肌酐、总胆固醇)含量及生化指标[总抗氧化能力(total antioxidant capacity,T-AOC)、超氧化物歧化酶(superoxide dismutase,SOD)活性、单胺氧化酶(monoamine oxidase,MAO)活性、B淋巴细胞瘤-2基因(B-cell lymphoma-2,Bcl-2)含量和半胱氨酸蛋白酶-3(Caspase-3)]活性。结果表明:分子量小于3 kDa的蛋白肽抗氧化活性最强。与模型对照组相比,南瓜籽蛋白肽中、高剂量组可改善衰老小鼠脏器系数,并使代谢产物(肌酐、总胆固醇)含量减少,同时南瓜籽蛋白肽高剂量组T-AOC、SOD活性和Bcl-2含量升高,MAO、Caspase-3的活性降低。说明南瓜籽蛋白肽可以有效改善衰老小鼠的脏器系数,抑制机体的氧化应激反应,调节组织细胞中凋亡因子活性,延缓衰老。

Abstract:This study aimed to explore the anti-aging effect of pumpkin seed protein peptides on D-galactoseinduced aging mice.In this study,the components with the strongest antioxidant activity were selected from pumpkin seed protein peptides with different molecular weights for the animal experiment.In the experiment,a normal control group,a model control group,and low-,medium-,and high-dose[250,500 mg/(kg·d)and 1 000 mg/(kg·d)]pumpkin seed protein peptides groups were set up.After 42 days of intervention by gavage,the body weight,organ coefficient,the content of metabolites(creatinine and total cholesterol),and biochemical indices[total antioxidant capacity(T-AOC),superoxide dismutase(SOD)activity,monoamine oxidase(MAO)activity,B-cell lymphoma-2(Bcl-2)content,and Caspase-3 activity]were measured.The experimental results showed that the protein peptide with a molecular weight of less than 3 kDa had the strongest antioxidant activity.Compared with the model control group,the medium-and high-dose pumpkin seed protein peptides groups could improve the organ coefficient of aging mice and reduce the content of metabolites creatinine and total cholesterol.At the same time,the high-dose pumpkin seed protein peptides group showed increased T-AOC,SOD activity,and Bcl-2 content and decreased activities of MAO and Caspase-3.The results showed that pumpkin seed protein peptides could effectively improve the organ coefficient of aging mice,inhibit the body's oxidative stress response,regulate the activity of apoptosis factors in tissue cells,and delay aging.
