中文摘要: 以丹凤牡丹花蕊为原料,采用超声波复合酶法提取,经过去色、去蛋白得到粗多糖。采用DEAE-52 纤维素柱层析纯化得到3 个水溶性多糖(water-soluble polysaccharide,WSP)组分(WSP-1、WSP-2、WSP-3),并采用葡聚糖凝胶G-100 凝胶色谱法对得率高且降糖作用好的多糖组分进一步纯化。结果表明:WSP-3 是分子质量为14 351 Da 的酸性均一多糖,残基同时包含了α 构型的糖苷键,具有三螺旋结构;扫描电镜观察WSP-3 呈大小不均一的棒状。抗氧化试验表明:WSP-3 对DPPH·、·OH 和ABTS+·具有一定的清除能力,IC50 分别为3.38、3.05、3.39 μg/mL。
Abstract:The crude polysaccharides were extracted from stamens of Paeonia suffruticosa by ultrasonic complex enzyme and obtained by decolorization and proteinization. Three water -soluble polysaccharide(WSP)components(WSP-1,WSP-2 and WSP-3)were purified by DEAE-52 cellulose column chromatography,and were further purified by Sephadex G-100 gel chromatography with high yield and good hypoglycemic activity.The results showed that WSP-3 was an acidic homozygous polysaccharide with a molecular weight of 14 351 Da,and the residues contained α-glycosidic bonds with a triple helix structure. WSP-3 was observed by scanning electron microscope as a rod of uneven size. Antioxidant experiments showed that WSP-3 had certain scavenging ability on DPPH·,·OH,and ABTS+·,and the IC50 was 3.38,3.05 and 3.39 μg/mL,respectively.
