中文摘要: 为提高黑青稞中β-葡聚糖的利用率,以6种菌种对带皮黑青稞进行单菌发酵及混菌复合发酵,筛选出最佳菌种及复配比例,并以β-葡聚糖得率为评价标准,通过单因素试验和响应面试验优化得到发酵富集青稞β-葡聚糖的最佳工艺,并对该工艺下得到的青稞β-葡聚糖进行体外抗氧化活性研究。结果表明:高活性干酵母、酿酒干酵母、嗜热链球菌在复配比 2∶1∶1(质量比),料液比 1∶12(g/mL)、接种量 5%(质量分数)、发酵温度 34℃、发酵时间 26 h时的β-葡聚糖得率最高,为(42.8±0.0)%。相比于富集前,富集后β-葡聚糖得率提高了63.01%,说明发酵富集青稞β-葡聚糖具有可行性。发酵富集得到的β-葡聚糖具有良好的抗氧化活性,在一定范围内,DPPH自由基清除率、ABTS+自由基清除率均能达到50%以上。
Abstract:This study aims to improve the utilization rate of β-glucan in black highland barley.To be specific,unhulled black highland barley was fermented with each of six strains to screen the optimal strains,and then the highland barley was fermented with the mixture of optimal strains at different ratios to obtain the optimal ratio.With the yield of β-glucan as the evaluation index,the fermentation conditions of highland barley were optimized by single factor test and response surface methodology.For the β-glucan obtained under the optimal conditions,the in vitro antioxidant activity was tested.The results showed that the highest yield([42.8±0.0)%]of β-glucan was achieved under the following conditions:mixture of highly-active dry yeast,alcohol-fermentation dry yeast,and Streptococcus thermophilus at the ratio of 2∶1∶1 (mass ratio),material-to-liquid ratio of 1∶12(g/mL),inoculum at 5% (mass fraction),temperature of 34 ℃,and fermentation for 26 h.The yield of β-glucan increased 63.01% after enrichment,indicating that it was feasible to enrich β-glucanin highland barley by fermentation.The yielded β-glucan had strong antioxidant activity and it can scavenge over 50% of the DPPH and ABTS+radical.
