甘蔗内生酵母菌的分离鉴定及发酵性能

中文摘要: 为研究甘蔗内生酵母菌的种类并筛选出发酵性能良好的酵母菌株,该文采用组织分离法对甘蔗中的内生酵母菌进行分离,通过形态学、生理生化以及26S rDNA D1/D2区序列分析法进行鉴定。利用杜氏管发酵法初筛产气性能较好的菌株,确定筛选菌株的最适生长条件,然后采用摇瓶发酵法对其进行耐糖、耐盐以及耐酒精等耐受性试验,复筛出发酵性能优良的菌株。结果表明,从甘蔗中共分离得到内生酵母菌63株,鉴定为8种:葡萄有孢汉逊酵母(Hanseniaspora uvarum)、季也蒙毕赤酵母(Meyerozyma guiliermondii)、汉逊德巴利酵母(Debaryomyces hansenii)、嗜油假丝酵母(Candida oleophila)、类筒假丝酵母(Candida zeylanoides)、异常威克汉姆酵母(Wickerhamomyces anomalus)、Cystobasidium minutum以及Papiliotrema terrestris。从63株酵母菌中,筛选一株生长性能和发酵性能优良的菌株J-6,在温度26℃、pH5.0的条件下,24 h时生长速率达到最大值,其最大值为1.993,J-6菌株可耐受体积分数20%乙醇、140 g/L氯化钠和300 g/L葡萄糖。在乙醇体积分数为16%、氯化钠60 g/L和葡萄糖150 g/L条件下,其OD600nm值最大,分别为1.935、1.608和1.901,经鉴定为异常威克汉姆酵母。
Abstract:The paper aimed at initially exploring the diversity of endophytic yeast in sugarcane and screening strains with good fermentation performance.The endophytic yeast in sugarcane was isolated by the tissue culture method,and then identified based on morphology,physiology,biochemistry,and 26S rDNA D1/D2 region analyses.Strains with good gas-producing performance were initially screened by the Duchenne tube fermentation method to determine their optimal growth conditions.Subsequently,the strains with excellent fermentation performance were further screened with sugar,salt,and alcohol tolerance tests by shaking-flask fermentation.Sixty-three strains of endophytic yeast were isolated and identified as eight species,namely Hanseniaspora uvarum,Meyerozyma guiliermondii,Debaryomyces hansenii,Candida oleophila,Candida zeylanoides,Wickerhamomyces anomalus,Cystobasidium minutum,and Papiliotrema terrestris.Among the above 63 strains,strain J-6 showed especially excellent growth and fermentation performance,whose growth rate reached the maximum(1.993)after 24 hours at 26 ℃ and pH5.0.It showed tolerance in the context of 20%ethanol,140 g/L sodium chloride,and 300 g/L glucose.Its OD600nmreached up to 1.935,1.608,and 1.901 under the conditions of 16%ethanol,60 g/L sodium chloride,and 150 g/L glucose,respectively,which was identified as W.anomalus.
