余甘子多酚纯化工艺研究及组分分析

中文摘要: 为纯化余甘子(Phyllanthus emblica L.)中的多酚类物质,该研究比较6种大孔树脂对余甘子多酚静态吸附-解吸能力,筛选出XAD-7HP树脂对余甘子多酚纯化效果最佳,并对其纯化工艺进行优化。结果表明,最佳纯化工艺为吸附时上样液浓度4 mg/mL、pH3、上样体积310 mL、上样液流速2 mL/min,解吸时乙醇体积分数95%、解吸液体积280 mL、解吸液流速2 mL/min,在此工艺条件下余甘子多酚纯度由16.45%提升到60.67%,经高效液相色谱法(high performance liquid chromatography,HPLC)分析后,冻干粉中没食子酸的含量为72.179 mg/g、柯里拉京的含量为35.443 mg/g、柯子林鞣酸的含量为42.731 mg/g、鞣花酸的含量为38.652 mg/g。

Abstract:This study aimed to purify polyphenols from Phyllanthus emblica L.The static adsorption and desorption capacities of six macroporous resins for polyphenols from P.emblica were studied and compared.XAD-7HP resin was selected as the most suitable resin for the purification of polyphenols from P.emblica,and the purification process was optimized.The results showed that the optimal purification process was as follows:the concentration of sample solution during adsorption 4 mg/mL,pH3,volume of sample solution 310 mL and flow rate 2 mL/min;the volume fraction of ethanol during desorption 95%,volume of desorption solution 280 mL and flow rate 2 mL/min.Under the above conditions,the purity of polyphenols from P.emblica was increased from 16.45%to 60.67%.After analysis by high performance liquid chromatography(HPLC),the content of gallic acid,corilagin,chebulinic acid and ellagic acid in freeze-dried powder was 72.179,35.443,42.731 mg/g and 38.652 mg/g,respectively.
