中文摘要: 为阐明籽瓜多糖对肠道益生菌生长的作用机制,该试验采用超声辅助水提法提取籽瓜多糖,用二乙氨乙基(DEAE)纤维素DE-52分离得到3个组分(SMP-1、SMP-2和SMP-3)被用来替代碳源,以比较它们对肠道益生菌的生长促进作用。然后对SMP-1进行分子量、单糖和官能团组成等结构表征分析。结果表明:SMP-1、SMP-2和SMP-3均能明显促进青春双歧杆菌、长双歧杆菌、婴儿双歧杆菌以及嗜酸乳杆菌的生长,且不同组分的促生长效果显著不同。其中,SMP-1的促进效果要优于SMP-2和SMP-3。SMP-1的分子量为97 403 Da,由阿拉伯糖和半乳糖二中单糖组成,物质的量百分比为 0.271% ∶0.729%,其主链连接方式为→6)-β-D-Galp-(1→3)-β-D-Galp-(1→的糖苷键,而端基α-L-Araf-(1→通过O-3键连接在主链上。
Abstract:To elucidate the mechanism of the growth-promoting effect of seed melon polysaccharides on intestinal probiotics,seed melon polysaccharides were extracted by ultrasound-assisted water extraction method.Subsequently,three fractions(SMP-1,SMP-2 and SMP-3)were obtained by DEAE cellulose DE-52 isolation.They were used as a substitute for carbon sources to compare their growth-promoting effect on intestinal probiotics.Further structural characterization of SMP-1 was carried out for molecular weight,monosaccharide,and functional group composition.The results showed that SMP-1,SMP-2 and SMP-3 significantly promoted the growth of Bifidobacterium adolescentis,Bifidobacterium longum,Bifidobacterium infantis,lactobacillus acidophilus and the growth-promoting effects of different fractions were significantly different.Among them,the growth-promoting effect of SMP-1 was better than that of SMP-2 and SMP-3.The molecular weight of SMP-1 was 97 403 kDa.Moreover,SMP-1 was primarily comprised of arabinose and galactose,and the molar ratio between them was 0.271∶0.729.The main chain of SMP-1 was connected by→6)-β-D-Galp-(1→3)-β-D-Galp-(1→ glycosidic bond,while the end group α-L-Araf-(1→ was attached to the main chain by an O-3 bond.
