柴达木大肥菇发酵过程中酚酸抗氧化活性变化

中文摘要: 采用液体发酵法发酵柴达木大肥菇,利用超声波辅助法提取胞内、胞外酚酸,通过测定菌丝体生物量、酚酸含量以及发酵过程中胞内、胞外酚酸的5种抗氧化指标,探究柴达木大肥菇发酵过程中酚酸抗氧化活性。结果表明:柴达木大肥菇菌丝体生物量和胞内、胞外酚酸产量达到最大时分别为(22.91±0.21)、(6.73±0.05)、(10.11±0.05)g/L,且胞外酚酸含量大于胞内酚酸含量;柴达木大肥菇胞内、胞外酚酸对不同自由基的清除能力不同,在发酵过程中抗氧化活性处于动态变化,且胞内活性高于胞外。胞内酚酸对2,2-联氮-二(3-乙基-苯并噻唑-6-磺酸)二铵盐阳离子[2,2'-azinobis-(3-ethylbenzthiazoline-6-sulphonate)cation,ABTS+]自由基、1,1-二苯基-2-三硝基苯肼(1,1-diphenyl-2-picrylhydrazyl,DPPH)自由基、超氧阴离子自由基、羟自由基的最高清除率及铁离子还原能力分别为44.52%、50.54%、63.90%、29.35%、7.17%;胞外酚酸对4种自由基的最高清除率及铁离子还原能力分别为47.32%、34.39%、23.95%、28.61%、3.67%。因此,柴达木大肥菇胞内、胞外酚酸是天然抗氧化剂的良好来源。
Abstract:In this paper,Agaricus bitorquis(Quél.)Sacc.was fermented by liquid fermentation ,and the intracellular and extracellular phenolic acid was extracted by ultrasonic-assisted method.The antioxidant activity of phenolic acids of A.bitorquis(Quél.)Sacc.during the fermentation was explored through the determination of mycelium biomass phenolic acid content,and the scavenging rate of intracellular and extracellular phenolic acids on 5 antioxidant indexes during fermentation.The results showed that the maximum mycelium biomass,intracellular phenolic acid content,and extracellular phenolic acid content were(22.91±0.21),(6.73±0.05)g/L and(10.11±0.05)g/L,respectively.The extracellular phenolic acid content was higher than the intracellular phenolic acid content.The intracellular and extracellular phenolic acids of A.bitorquis (Quél.)Sacc.had different scavenging abilities to different free radicals.The antioxidative activity was in a dynamic state during the fermentation,and the intracellular activity was higher than extracellular activity.The highest scavenging rate of intracellular phenolic acids to 2,2'-azinobis-(3-ethylbenzthiazoline-6-sulphonate)cation(ABTS+)radical,1,1-diphenyl-2-picrylhydrazyl(DPPH)radical,superoxide anion radical,and hydroxyl radical,and the iron ion reduction ability were 44.52%,50.54%,63.90%,29.35% and 7.17%,respectively.The highest scavenging rate and the iron ion reduction ability of extracellular phenolic acids were 47.32%,34.39%,23.95%,28.61% and 3.67%,respectively.Therefore,intracellular and extracellular phenolic acids of A.bitorquis(Quél.)Sacc.are good sources of natural antioxidants.
