中文摘要: 为提高辣木籽异硫氰酸酯的稳定性及生物利用率,使用薄膜分散法,以卵磷脂和胆固醇的质量比、卵磷脂和胆酸钠的质量比、超声时间为影响因素,以包封率为评判标准,采用单因素和响应曲面法进行辣木籽异硫氰酸酯柔性脂质体的工艺优化。使用低温超速离心法分离柔性脂质体中游离的辣木籽异硫氰酸酯以计算其包封率;通过紫外分光光度计法测定异硫氰酸酯含量。结果表明,辣木籽异硫氰酸酯柔性脂质体优化后的工艺为卵磷脂与胆固醇质量比9.95∶1、卵磷脂与胆酸钠质量比10.32∶1、超声时间7.78 min,在此条件下制备的辣木籽异硫氰酸酯柔性脂质体包封率为64.17%。通过表征,辣木籽异硫氰酸酯柔性脂质体的粒径为348 nm,多分散系数为0.251,Zeta电位为-24.7mV。透射电镜显示该柔性脂质体分散比较均匀,呈较规则圆形。经过24 h体外释放率检测后,发现该柔性脂质体体外释放率最后保持在80.105%,具有缓释的效果。
Abstract:This study aimed to improve the stability and bioavailability of isothiocyanate in Moringa oleifera seeds.To be specific,the flexible liposome was prepared with the thin film dispersion method.With the evaluation index of entrapment efficiency,the mass ratio of lecithin to cholesterol,mass ratio of lecithin to sodium cholate,and ultrasonic time in the preparation of the flexible liposome of M.oleifera seed isothiocyanate were optimized by single factor test and response surface methodology.The free isothiocyanate in liposomes was separated by low-temperature ultracentrifugation,and isothiocyanate content was determined by UV spectrophotometry.The results showed that the optimized process was as follows:lecithin-to-cholesterol mass ratio of 9.95∶1,lecithin-to-sodium cholate mass ratio of 10.32∶1,and ultrasonic time of 7.78 min.Under these conditions,the entrapment efficiency was calculated to be 64.17%.Moreover,the yielded isothiocyanate-loaded liposomes had the particle size of 348 nm,polydispersity index(PDI)of 0.251,and the Zeta potential of-24.7 mV.Transmission electron microscopy(TEM)showed that the liposomes were fairly uniform and regular round.After 24 h,the in vitro release rate of the liposomes was 80.105%,marking the ideal slow-release effect of the liposomes.
