中文摘要: 以葛根渣为主要原料,按渣麸质量比8∶3添加麸皮,再加入固体成分质量70%的水分及适量无机盐组成培养基,30℃下进行黑曲霉固态发酵。利用发酵过程中产生的木聚糖酶和羧甲基纤维素(caboxy methyl cellulose,CMC)酶对葛根细胞壁纤维组织的降解破坏作用,促进葛根素由胞内溶出。结果表明,当分别发酵至72 h和96 h时,木聚糖酶和CMC酶可分别测出最大活力,当发酵至120 h时,测得发酵物料中葛根素含量接近最大值,经高效液相色谱(high performance liquid chromatography,HPLC)法检测得5.68 mg/g,比直接醇提葛根渣获得的葛根素含量提高了74.8%,实现了葛根素的高效制备。
Abstract:Pueraria root residue is a bulk waste obtained after processing Pueraria roots,and it is rich in the biologically active ingredient puerarin.The medium was obtained using Pueraria root residue as the main raw material,adding bran at a ratio of 8∶3 (ω/ω)and then adding 70% of the solid mass of water and an appropriate amount of inorganic salt.The solid-state fermentation was conducted at 30°C using Aspergillus niger.The degradation of the fibrous tissue of the Puerariacell wall by xylanase and CMC enzyme promotes the release of puerarin from the cell.The results showed that the maximum activity of xylanase and CMCase could be measured separately when fermented for 72 h and 96 h,respectively.When fermented for 120 h,the puerarin content in the fermented material was close to the maximum value determined through high performance liquid chromatography,which was 5.68 mg/g.Compared with the direct alcohol extraction of the Pueraria root residue,the content of puerarin obtained increased by 74.8%,achieving an efficient puerarin preparation.
