不同生长年份桑黄中活性成分动态分析

中文摘要: 以同一桑黄菌种为研究对象,通过与野生桑黄进行比较,分析桑黄菌丝体、一年生、两年生、三年生和四年生桑黄子实体中活性成分的差异。采用苯酚-硫酸法、福林酚法对不同桑黄提取物中的总多糖(水提物)和总多酚(醇提物)含量进行评价,进一步应用液相色谱-质谱联用(liquid chromatograph-mass spectrometer,LC-MS)分析比较桑黄醇提物中主要活性成分。结果表明,一年生桑黄中总多糖含量较高,而四年生桑黄中总多酚含量较高;通过LC-MS分析发现,桑黄菌丝体中活性成分较少,与总多酚含量测定结果相符,人工栽培桑黄和野生桑黄的醇提物成分较为相近,野生桑黄中成分种类较多,但人工栽培桑黄中个别成分高于野生桑黄;通过LC-MS技术初步鉴定出桑黄提取物中6种化合物,分别为 phellinin B、phelligridimer A、davallialactone、phellinstatin、hypholomine B 和 inoscavinA。
Abstract:The differences in active components of annual,biennial,triennial and quadrennial fruiting bodies of Phellinus igniarius were analyzed in comparison with wild P.igniarius.The contents of polysaccharides(water extract)and polyphenols (alcohol extract)in different extracts of P.igniarius were evaluated using the phenolsulfuric acid and folin-phenol methods,and the main active components in the alcohol extract of P.igniarius were analyzed and compared using liquid chromatography-mass spectrometry (LC-MS).The results showed that the content of polysaccharide was higher in biennial P.igniarius,while that of polyphenols was higher in quadrennial P.igniarius.The results of LC-MS analysis showed that there were fewer active components in the mycelium of P.igniarius,which was consistent with the results of polyphenol content determination.In addition,the alcohol extracts of artificial and wild P.igniarius were similar.There were more components in the wild P.igniarius,but some components in the artificial were much higher than those in the wild.Six compounds were identified by LC-MS,namely phelinin B,phelligridimer A,davallialactone,phellinstatin,hypholomine B,and inoscavin A.
